Abstract
Bu çalışmada, brusella antijeni ve anti-rabbit IgG antikoru kullanılarak PVC ve PVC-NH2 de temel teşkil eden brusella ve anti-rabit IgG potansiyometrik immünosensörler hazırlandı. Hazırlanan sensörlerin potansiyometrik performansları bilgisayar kontrollü potansiyometrik sistem kullanılarak test edildi. PVC temelli brusella immünosensör 3,125-50 ng/mL konsantrasyon aralığında anti-brusellaya doğrusal davranış sergilerken (R2 = 0,993), PVC-NH2 temelli brusella immünosensör 6,25-100 ng/mL konsantrasyon aralığında anti-brusellaya doğrusal davranış sergilemekteydi (R2 = 0,997). PVC ve PVC-NH2 anti rabbit IgG immünosensörlerin her ikisi de 25-200 ng/mL konsantrasyon aralığında rabit IgG ye karşı doğrusal davranış sergilemekteydi (R2 = 0,995 her iki immünosensör için). Bütün immünosensörlerin cevap zamanı oldukça uzundu. Genel olarak konsantrasyon arttıkça cevap zamanında artış gözlemlendi. Geliştirilen immünosensörlerle bir örnek çözeltiden 3-5 kez tekrarlanabilir potansiyometrik sinyal elde edildi.
In the present study, Brucella and anti-rabit IgG potentiometric immunosensors based on PVC and PVC-NH2 were prepared by using brucella antigen and anti-rabit IgG antibody. Potentiometric performances of the immunosensors prepared were tested using a computer controlled potentiometric system. Brucella immunosensor based on PVC-NH2 exhibited a linear response between 6,25-100 ng/mL anti-brucella concentrations (R2 = 0,993) while brucella immunosensor based on PVC exhibited a linear response between 3,125-50 ng/mL anti-brucella concentrations (R2 = 0,997). Anti-rabit IgG immunosensors based on PVC and PVC-NH2 both produced a linear response with R2 = 0,995 between 25-200 ng/mL rabbit IgG concentartions. The responses of the immunosensors used were found highly time consuming. In general, the response time of the immunosensors increased with the increase of sample concentration. So far, for a sample solution, the developed immunosensors exhibited reproducible signals up to 3-5 measurements
