Abstract
Bu çalışmada, Ters-Faz Yüksek Basınç Sıvı Kromatografisi (HPLC) kullanılarak fenolik bileşiklerin(fenol, m-krezol ve rezorsinol) tayinleri için bir yöntem geliştirildi. Bunun için bazik ortamda peroksidisülfat varlığında 4-aminoantipirin ile türevlendirilerek UV-Gör. aktif türevlerine dönüştürülen fenolik bileşikler kloroform ile sulu fazdan ekstrakte edildi. Ekstrakte edilen UV-Gör aktif fenolik türevler, HPLC sistemine enjekte edilerek asetonitril-su (%45:55 v/v) hareketli faz kompozisyonu ile ters faz C18 kolonunda ayırıma tabi tutuldu ve 440 nm dalga boyuna ayarlı spektrofotometrik dedektör ile tayin edildi. Bu üç fenol için tayin süresi 14 dakikadan az ve hesaplanan tayin sınırları 50 ?L su numunesi kullanıldığında 0,015 -3,666 ?g/L arasında bulundu. Geliştirilen metod baraj ve nehir suyu örneklerine uygulandı. Baraj ve nehir suyundaki fenol ve m-krezol'ün ortalama konsantrasyonları 0,033 ?g/L iken, rezorsinolün ortalama konsantrasyonu 12,935 mg/L olarak belirlendi.Anahtar Kelimeler: Fenolik Bileşikler, HPLC, Derivatizasyon, Aminoantipirin
In this study, a method was developed for the indications of compounds of phenol (pHenol, m-cresol and resorcinol ) by using high pressure liquid cyromotograpHiy (HPLC). For this study, phenolic compounds were extracted with the chloroform which were the phenolic compounds transformed into UV-Vis active derivatives that were derivatiated by 4-aminoantipyrine in the existence of peroxydisulphate in alkaline medium. Extracted UV-Vis active phenolic derivatives, injected to HPLC system, were dissociated with acetonitrile-water (%45:55 v/v) mobile phase composition in reversed- faz C18 column and they were indicated with the spectrophotometric dedector adjusted to 440 nm wavelength. For these three phenols, retention time of indication was less than 14 minutes and limits of detection was found between 0,015 -3,666 µg/Lwhen 50 µL water samples was used. The developed method was applied to the dam and river water sample. The phenol in barrage and river water, rezorsinolun average concentration was determined as 12,935 mg/L when the average concentrations of m-krezol was 0,033 µg/L.Key Words: Phenolic compounds, Derivatization, HPLC, Aminoantipyrine
