Abstract
Dünyada yaklaşık 300 Polygonum türü bulunmaktadır. Türkiye` de Polygonum türüne ait 45 taksonu vardır ve 13 tanesi endemiktir. Son zamanlarda, literatürde Polygonum türleri ile ilgili biyoaktif moleküllerin izolasyonu, karakterizasyonu, antioksidan, antimikrobial, antitümör, antiobozite gibi biyolojik aktivitelerinin araştırıldığına rastlanmaktadır. Dünyada Polygonum türleri ile ilgili biyokimyasal özelliği oldukça fazla araştırılmasına karşılık Endemik Polygonum samsunicum L.` nin kimyasal içerikleri, enzim inhibisyon potansiyeli, antioksidan ve antiproliferatif aktiviteleri ile ilgili herhangi bir çalışma bulunmamaktadır. Tez çalışmasında P. samsunicum (M) su (MSU), etanol (MET), metanol (MME), aseton (MAS), etilasetat (MEA) ve hekzan (MHE) ile ekstrakte edildi. Ekstraktların kimyasal içerikleri (fenolik bileşenleri, toplam fenolik ve flavonoid içeriği), antioksidan (toplam antioksidan aktivitesi, indirgeme gücü, lipit peroksidasyon inhibisyonu, metal şelat, deoksiriboz koruma, hidrojen peroksit, süperoksit anyonu, serbest radikal, hidroksil radikali ve ABTS˙⁺ radikali giderme), antiproliferatif, asetilkolin esteraz ve bütirilkolin esteraz inhibisyon aktiviteleri spektroskopik olarak belirlendi. MME ekstraktının ekstraktsiyon verimi diğer ekstraktlara göre daha yüksek (%13.64) bulunmuştur. Ekstraktların toplam fenol ve flavonoid içeriklerinin yüksek oranda olduğu tespit edildi. MME` nin toplam fenolik içeriği (16.10±0.15 mg gallik asit eşdeğer/g kuru bitki) ve MEA` nın toplam flavonoid içeriği (17.40±0.68 mg kateşin eşdeğer/g kuru bitki) en yüksek olduğu bulundu. Klorojenik asit, kateşin, ellagik asit, kuersetin-3-B-D-glukosit, kihikorik asit, naringin, morin ve sinnamik asit bileşikleri MET, MME, MHE, MAS, MEA ve MSU ekstraktlarında yüksek oranda belirlenmiştir. MSU ve MME ekstraktlarının toplam antioksidan aktivitelerinin yüksek olduğu belirlendi ve fenoliklerin analiz sonuçlarını desteklemektedir. Ekstraktların in vitro antioksidan aktiviteleri artan konsantrasyona bağlı (50-500 μg/mL) artış olduğu belirlendi (p<0.05). MSU ekstraktının lipit peroksidasyonu inhibisyonu (EC50= 12.51±4.22 μg/mL), MME ekstraktının indirgeme gücü kapasitesi (A0.5= 101.9±3.91 μg/mL), MME ekstraktının H2O2 giderme aktivitesi (EC50= 26.94±4.04 μg/mL), MSU ekstraktının deoksiriboz koruma aktivitesi (EC50= 41.48±2.44 μg/mL), MSU ekstraktının süperoksit anyonu giderme aktivitesi (EC50= 15.43±2.92 μg/mL) diğer ekstraktlara göre daha etkin aktiviteler göstermiştir. Ekstraktların asetilkolin ve bütirilkolin esteraz inhibisyon potansiyelleri galantamine göre daha etkili olduğu bulunmuştur. MME, MET, MAS ve MHE ekstraktları HeLa kanser hücre hatlarına karşı yüksek antiproliferatif aktivite gösterdi. Sonuç olarak, P. samsunicum` un MSU, MET, MME, MAS, MEA ve MHE ekstraktlarının kimyasal analizleri ve biyokimyasal sonuçları potansiyel antioksidan, antinödejeneratif ve antikanser doğal ürün olabildiğini desteklemektedir. Recently, it has been observed in the literature that the isolation, characterization, antioxidant, antimicrobial, antitumor, and antiobosity biological activities of Polygonum species have been investigated. Although the biochemical properties of Polygonum species have been investigated in the world, there are no studies on the chemical contents, enzyme inhibition potential, antioxidant and antiproliferative activities of Endemic Polygonum samsunicum L. In the thesis study, P. samsunicum (M) was extracted with water (MSU), ethanol (MET), methanol (MME), acetone (MAS), ethylacetate (MEA) and hexane (MHE). The chemical contents (phenolic components, total phenolic and flavonoid content), antioxidan (total antioxidant activity, reducing power, inhibition of lipit peroxidation, metal chelate, deoxyribose protection, hydrogen peroxide, superoxide anion radical, free radical, hydroxyl radical and ABTS˙⁺ radical removal), antiproliferative, acetylcholinesterase and butylcholinesterase inhibition activities of extracts were determined spectroscopically. The extraction yield of MME extract (13.64 %) was higher than other extracts. The total phenol and flavonoid contents of the extracts were found to be high. The total phenolic content of MME (16.10±0.15 mg gallic acid equivalent/g dry plant) and the total flavonoid content of MEA (17.40±0.68 mg catechin equivalent/g dry plant) were highest. Chlorogenic acid, catechin, elllagic acid, quercetin-3-B-D-glucoside, chichoric acid, naringin, morin and cinnamic acid compounds were highly identified in MET, MME, MHE, MAS, MEA and MSU extracts. The total antioxidant activities of MSU and MME extracts were found to be high and support the analysis results of phenolics. The in vitro antioxidant activities of the extracts were determined to be an increase due to increasing concentration (50-500 μg/mL) (p<0.05). Inhibition of lipit peroxidation of MSU extract (EC50= 12.51±4.22 μg/mL), reducing power capacity of MME extract (A0.5= 101.9 ± 3.91 μg/ mL), H2O2 removal activity of MME extract (EC50= 26.94±4.04 μg/mL), deoxyribose protection activity of MSU extract (EC50= 41.48±2.44 μg/mL) and superoxide anion removal activity of MSU extract (EC50= 15.43±2.92 μg/mL) showed more effective activities than the other extracts. In conclusion, the chemical analyzes and biochemical results of P. samsunicum extracts of MSU, MET, MME, MAS, MEA and MHE support potential antioxidant, antineodegenerative and anticancer natural products.
