Abstract
Çalışmada, kistik fibrozis tanısında kullanılmak üzere 4-ter-Butilkaliks[4]aren-tetraasetik asit tetraetil ester (sodyum iyonofor-X) iyonoforu kullanılarak mikro büyüklükte Na+-seçici elektrot, mezo-tetrafenilporfirinmanganez (III)(klorür iyonofor-I) kullanılarak mikro büyüklükte Cl- seçici elektrotlar geliştirildi. Geliştirilen elektrotların potansiyometrik performansları durgun ve hareketli ortamlarda bilgisayar kontrollü potansiyometrik bir sistem kullanılarak test edildi. Elektrotların durgun ve hareketli ortam ölçümlerinden tekrarlanabilirliklerinin yüksek olduğu ve 10-1-10-5 M konsantrasyon aralığında doğrusal cevap sergilediği gözlendi. Geliştirilen elektrotların hareketli ortam ölçüm hücreleri kullanılarak ter klorür ve sodyum düzeyleri ölçüldü. Elde edilen veriler Ondokuz Mayıs Üniversitesi Tıp Fakültesinde uzun süreden beri kullanılan Wescor model kistik fibrosis ölçüm sistemiyle elde edilen verilerle karşılaştırmalı olarak incelendi. Elde edilen sonuçların hastanedeki ölçüm değerleriyle uyum içerisinde olduğu görüldü. Geliştirilen elektrotların kistik fibrozis hastalığı tanısında etkin olarak kullanılmasını sağlayan bir ?kistik fibrozis tanı sistemi? geliştirildi. Kistik fibrozis tanı sistemi, geliştirilen mikro elektrot ve iyontoforez elektrot sistemlerini içermektedir. İyontoforez sistemde pilokarpin maddesi, acısız olarak deri altına gönderilerek o bölgede terlemenin oluşmasını sağlamaktadır. Oluşan ter deri yüzeyine çıkıp birikmeye başladığında, akış yolunu takip ederek hareketli ortam hücresindeki elektrotlara yönelmektedir. Elektrotlara yönelen terin içindeki klorür ve sodyum düzeyleri elktrotlar tarafından eşzamanlı olarak ölçülmektedir.
In the present study, micro sized Na+ selective electrode based on ionophore 4-tert-Butylcalix[4]arene-tetraacetic acid tetraethyl ester (sodium ionophore-X) and micro sized Cl- selective electrode based on meso-Tetraphenylporphyrin manganese(III)-chloride complex (chloride ionophore-I) were developed for the use of cystic fibrosis dignosis. Potentiometric performance characteristics of the developed micro sized electrodes were tested under static and dynamic conditions by using a computer-controlled potentiometric system. The micro-sized electrodes performed linear response over the concentration range of 10-1-10-5 M and their reproducibilities were also quite well. Chloride and sodium levels of sweat samples supplied from the hospital of Ondokuz Mayis University were measured by using flow through cells of the developed micro-sized electrodes. The obtained data was comperatively examined with the data received by the Wescor model measuring system that was in use for long time in the hospital. It can be concluded that the data obtained from the micro-sized electrodes was in agreement with the data obtained from the hospital. A novel ?cystic fibrosis diognosis system? has consequently been developed for the diognosis of cystic fibrosis desease by providing efficient use of the micro-sized electrodes developed. The cystic fibrosis diognosis system was consisted of iontophoresis cell and micro-electrode cell systems. In the iontophoresis cell system, the pilocarpine material, forced into the skin without any pain by the electrode placed in the cathode, causes sweating over the area of the skin. When sweat reaches and coagulates onto the surface of the skin, then starts streaming toward the micro-electrode cell following the flow-path in the system. The chloride and sodium levels in the sweat passed the micro-electrode cell are simultaneously measured by the micro-sized electrodes.
