ÖZET Yüksek Lisans Tezi ORDU İLİNDE ELMA AĞAÇLARINDA ATEŞ YANIKLIĞI HASTALIĞINA NEDEN OLAN ERWINIA AMYLOVORA ETMENİNİN İZOLASYONU, TANISI VE YAYGINLIĞININ BELİRLENMESİ Süreyya Uzer Ondokuz Mayıs Üniversitesi Fen Bilimleri Enstitüsü Bitki Koruma Anabilim Dalı Danışman: Doç. Dr. Hasan Murat Aksoy Bu çalışma; 2015 - 2016 yılları arasında ateş yanıklığına sebep olan Erwinia amylovora'nın Ordu ilindeki yaygınlık durumu, 6 ilçede survey yapılarak belirlenmiştir. Bakteriyel izolatlar, biyokimyasal olarak; Nutrient glukoz agar ve King B besiyerlerinde gelişim, oksidatif - fermentatif test, 36 °C'de gelişim, KOH testi, katalaz testi, jelatinin sıvılaştırılması, D(+) Mannose, D(+) Trehalose, Rafinose D(+) Xylose, D(-) Arabinose, L(+) Rhamnose, sorbitol' den asit üretimi, tütünde aşırı duyarlılık ve patojenisite testlerine göre değerlendirilmiştir. İzolatların moleküler tanısında ise touchdown koloni PCR yöntemine göre yapılmıştır. 2015 - 2016 yıllarının ortalama hastalık bulunma oranları sırasıyla; Merkez % 32.85, Fatsa % 28.12, Ulubey % 26.12 Korgan % 23.20, Perşembe % 20.75 ve Gülyalı % 15.36, hastalık yaygınlık oranları ise sırasıyla Ulubey % 85, Fatsa % 78, Merkez % 72.5, Gülyalı % 71.5, Perşembe % 71 ve Korgan % 67.5 olarak belirlenmiştir. ABSTRACT Master's Thesis ISOLATION, IDENTIFICATION AND DETERMINATION OF THE PREVALENCE OF ERWINIA AMYLOVORA CAUSING FIRE BLIGHT IN APPLE TREES IN ORDU PROVINCE Süreyya Uzer Ondokuz Mayıs University Graduate School of Natural and Applied Sciences Department of Plant Protection Supervisor : Assoc. Prof. Dr. Hasan Murat Aksoy This study was performed to determine the incidence of fire blight caused by Erwinia amylovora in 6 districts Ordu Province in 2015 - 2016 years. In identification of the bacterical isolates, growth on Nutrient glucose agar and King's B media, biochemical; oxidative - fermentative test, growth at 36 °C, KOH tests, catalase tests, gelatin liquefaction, acid production from, D(+) Mannose, D(+) Trehalose, Rafinose D(+) Xylose, D(-) Arabinose, L(+) Rhamnose, Sorbitol, hypersensitive reaction on tobacco tests and pathogenicity were based. Molecular identification of the isolates were done by touchdown colony PCR method. These agent certainty of presence in Ordu province was revealed for the first time in this performed diagnostic study. The mean rates of disease and the disease prevalence was found to be in Merkez 32.85%, Fatsa 28.12 %, Ulubey 26.12% Korgan 23.20%, Perşembe 20.75% and Gülyalı 15.36 % Ulubey 85%, Fatsa 78%, Merkez 72.5%, Gülyalı 71.5%, Perşembe 71% and Korgan 67.5% restpectively in 2015 - 2016 years.
