Abstract
Apetala2-ethylene-responsive element binding factor (AP2-ERF) süperailesi gelişimsel ve fizyolojik olarak önemli rollere sahiptir. Fasulye genomunun dizilemesi tamamlandığından dolayı AP2-ERF genlerinin hepsinin tanımlanması mümkün hale gelmiştir. Bu çalışmada biyoinformatik analizler ile 180 AP2-ERF süperailesi geni bulunmuş ve bu genlerden PvAP2-ERF31 geninin karakterizasyonu yapılmıştır. PvAP2-ERF gen ailesinin yerel hizalama ve filogenetik analizleri, intron/exon yapısı, genlerinin kromozomlar üzerindeki dağılımı ve motif tahmini yapılmıştır. Ayrıca PvAP2-ERF genlerinin promotor yapıları ve miRNA hedefleri, gen ontolojisi ve fonksiyonel anlamlandırma işlemleri gerçekleştirilmiştir. AP2-ERF proteinleri arasındaki etkileşimler, amino asitlerin fiziksel ve kimyasal karakteristikleri analiz edilmiştir. Ek olarak PvAP2-ERF genlerinin ekspresyo seviyeleri in silico ve qRT-PCR analizi ile değerlendirilmiştir. Biyoinformatik analizler sonucunda PvAP2-ERF31 geni Gateway klonlama sistemiyle klonlanıp Agrobacterium ile tütüne aktarılmıştır. T1 Transgenik tütün adaylarının moleküler ve fizyolojik analizler yapılmıştır. T1 transgenik adaylar 200 mM NaCl içeren ortamlarda stres testine tabi tutulmuş ve transgenik tütün adayları sağlıklı bir şekilde çimlenmiş ve büyümüştür.

Since the sequencing of the bean genome is complete, it has become possible to identify all of the AP2-ERF genes. In this study, 180 AP2-ERF superfamily genes by bioinformatic analysis were identified and among these genes, PvAP2-ERF31 gene was characterized. Distribution of genes on chromosomes, intron / exon structure, local alignment and phylogenetic analysis of the PvAP2-ERF gene family and motif prediction were done. In addition, gene ontology and functional annotation, promoter structures and miRNA targets of PvAP2-ERF genes were performed. Interactions between AP2-ERF proteins, physical and chemical characteristics of amino acids have been analyzed. In addition, the expression levels of PvAP2-ERF genes were assessed by in silico and qRT-PCR analysis. As a result of bioinformatics analysis, PvAP2-ERF31 gene was cloned by Gateway cloning system and transferred to Agrobacterium to tobacco. Molecular and physiological analyzes of T1 transgenic tobacco candidates were carry out. T1 transgenic candidates were done stress test in medium containing 200 mM NaCl and transgenic tobacco candidates were germinate and grown up as a healty.
