Publication: Plasenta Previa Hastalarında Kisspeptin İfade Düzeylerinin Değerlendirilmesi
Abstract
Plasenta previa etiyolojisinde kisspeptin düzeylerinin olası etkisinin tespiti, hamileliğin erken dönemlerinde plasenta previa tanı ve teşhisinde önemli bir rol oynayabilir. Bu araştırmada, plasenta previa tanısı alan hastalar ile normal gebelikler arasında kisspeptin ifade düzeylerindeki olası değişikliklerin tespit edilmesi amaçlanmıştır. Hastalar ve Yöntem Ondokuz Mayıs Üniversitesi Kadın Hastalıkları ve Doğum Perinatoloji polikliniğinde 2021-2022 yılları arasında plasenta previa tanısı almış gebeler ve normal gebeler çalışmaya dahil edildi. Gravida, parite, gebelik haftası, anne yaşı ve plesantanın yeri değerlendirildi. Plazma kisspeptin seviyeleri biyokimyasal olarak, plasenta kisspeptin ve kisspeptin reseptörü gen ifade düzeyi genetik olarak, plasenta kisspeptin düzeyi immünohistokimyasal olarak tespit edildi. Bulgular Serumda KISS1 konsantrasyon düzeyinin değerlendirmesi sonucunda, kontrol grubunda plasenta previa grubuna oranla belirgin bir artış ve istatiksel olarak ileri düzeyde anlamlı bir fark gözlenmiştir (p<0,001). Kontrol ve plasenta previa grupları arasında gerçekleştirilen istatiksel değerlendirme neticesinde gruplar arasında istatiksel olarak anlamlı bir farklılık saptanmıştır (p<0,05). Kontrol grubunda KISS1R gen ifade düzeyinin belirgin bir şekilde daha yüksek olduğu gözlenmiştir. Tartışma ve Sonuç Plasenta previalı hastalarda kontol grubundaki hastalara göre kisspeptin ifade düzeyleri daha düşük çıkmıştır. Hem biyokimyasal ve immünohistokimyasal hem de genetik analizler neticesinde alınan bulgular, plasenta previalı hastalarda kisspeptin ifade düzeyinin ciddi oranda daha düşük olduğunu göstermiştir. Detection of the possible effect of kisspeptin levels on the etiology of placenta previa may play a crucial role in the diagnosis of the placenta previa in the early stages of pregnancy. In this study, we aimed to determine possible changes in kisspeptin expression levels between patients diagnosed with placenta previa and normal pregnancies. Materials and Methods Pregnant and normal pregnant women who were diagnosed with placenta previa in Ondokuz Mayıs University Gynecology and Obstetrics Perinatology outpatient clinic between 2021-2022 were included in the study. Gravida, parity, gestational week, maternal age and placenta location were evaluated. Plasma kisspeptin levels were determined biochemically, placenta kisspeptin and kisspeptin receptor gene expression levels were determined genetically, and placenta kisspeptin levels were determined immunohistochemically. Results As a result of the evaluation of the KISS1 concentration level in the serum, a significant increase in the control group compared to the placenta previa group and a statistically significant difference was observed (p<0.001). As a result of the statistical evaluation performed between the control and placenta previa groups, a statistically significant difference was found between the groups (p<0.05). It was observed that the KISS1R gene expression level was significantly higher in the control group. Conclusions In our current study, kisspeptin expression levels were found to be lower in patients with placenta previa compared to patients in the control group. Findings obtained as a result of both biochemical, immunohistochemical and genetic analyzes showed that kisspeptin expression level was significantly lower in patients with placenta previa.
