Abstract
Xanthomonas campestris pv. campestris (Pammel) Dowson dünya genelinde brassica bitkilerinin en önemli bakteriyel hastalığı olarak bilinen siyah damar çürüklüğüne neden olan gram negatif bir bakteridir. Ekonomik öneme sahip lahanagil bitkilerinde etmenin en tipik belirtisi yaprak kenarlarında V-şeklinde sarı lezyonlara ve damarlarda siyahlaşmaya neden olmasıdır. Bu çalışmada 2016 - 2017 yılları arasında kış döneminde Bafra ovasında karnabahar yetiştirilen alanlarda tipik belirti gösteren hastalıklı bitki örnekleri toplanarak bakteriyel izolasyon yapılmıştır. NGA besi yerinde gelişen sarı renkli 110 izolatın 27 adedi tütünde aşırı duyarlılık ve karnabahar bitkisinde patojenisite testine pozitif reaksiyon göstermiştir. Biyokimyasal, patojenisite, 16S rDNA and gryB gene dizi analizi sonucunda patojen izolatların X. campestris pv. campestris olduğu belirlenmiştir. X. campestris pv. campestris'e ait KP19, KS27, BD14 ve BT13 isimli izolatlar, karnabahar bitkisinde Türkiye'de ilk kayıt olarak uluslararası indekse giren dergide yayımlanmıştır. Xanthomonas campestris pv. campestris (Pammel) Dowson is the one of the important bacterial disease known black rot in Brassicaceae family, a Gram-negative, bacterium. The most tipically symptoms of this disease is V-shaped necrosis in leaf edge in economical important crucifers. This thesis is a study to determine bacterial disease factors in cauliflower plants in Bafra Plain, Samsun Province. During the winter season between 2016 and 2017, diseased plant samples were collected which were similar to the symptoms of bacterial disease for the study. Twentyseven of one hundred and ten isolates growth yellow colony on NGA showed hypersensitivity reactions in tobacco and positive reaction for pathogenicity test. Biochemical, pathogenicity, 16S rRNA and grB gene sequence analysis were performed. As a result of these analyses isolates were identified as Xanthomonas campestris pv. campestris. X. campestris pv. campestris isolates KP19, KS27, BD14 and BT13 as the first record of the cauliflower plants in Turkey has been published in journals in the international index.
