Atrisorb Uygulanmıs ve Radikal Süpürücü Enzimler (Superoxide dismutase(SOD) ve Catalase (CAT)) Verilmis Köpeklerin Diseti Dokularında RadikalSüpürücü Enzimlerin (SOD, CAT ve Glutathione Peroxidase) AktivitelerininZamana Göre DegisimiÖZETBir rejeneratif tedavi yöntemi olan ?Yönlendirilmis Doku Rejenerasyonu? (YDR)isleminde, periodontal hastalık sonucu kaybedilmis periodontal dokuların rejenerasyonuamacıyla polilaktik asit içerikli bir bariyer olan Atrisorb membranı kullanılmaktadır.Daha önceki arastırmalar sonucunda, yabancı madde uygulanmasına,iskemireperfuzyona,inflamasyona ve baska faktörlere baglı olarak yara iyilesmesi sürecindeserbest oksijen radikalleri türevlerinin (SORT) olustugu verilerine dayanarak, buçalısmada dokuda meydana gelebilecek muhtemel yan etkileri azaltmak için lipozomiçine hapsedilmis SOD ve CAT kullanılmıstır. Daha sonra bu dokularda SORT'ungöstergesi olarak düsündügümüz radikal süpürücü enzimlerden (RSE) SOD, CAT veGlutathione peroxidase (GSH-Px)'nin 1 ay süre boyunca aralıklı olarak aktiviteleriölçülerek bölgede meydana gelen SORT miktarı konusunda kontrol ve test gruplarındakarsılastırmalı olarak analiz yapılmıstır.Çalısmada cinsiyet ayrımı yapılmaksızın sistemik ve periodontal açıdan saglıklı 7adet yetiskin köpek (10-15 kg) tercih edilmistir. Hayvanlar ticari olarak satılan yumusakköpek maması ile beslenilmis ve herhangi ilaçla muamele olunmamıslar. Bakımkompleksinde bakıma alınan hayvanlar defekt olusumu ve stabilizasyon periyotlarısonrasında Periodontolog denetiminde rejeneratif tedavi islemlerine tabi tutulmustur.Operasyon öncesinde ve operasyon sonrası tedavi süresi boyunca her 3 günde birolmaküzere bag dokusu ve epitel dokusu karısımı 0,5 x 0,5 cm boyutunda diseti biyopsiörnekleri alınmıstır. Bu örnekler, enzim aktivite tayini yapılıncaya kadar, -70 ºC' de ve0,25 M' lık sükroz çözeltisi içinde saklanmıstır.Enzim aktiviteleri belirleme islemlerine baslamadan önce, dokular homojenizasyon,freeze-thawing (dondurma-eritme), sonifikasyon ve santrifugasyon islemlerine tabitutulmus ve dokulardan supernatant elde edilmistir. Elde edilen supernatantta McCordve Fridovich, Flohe ve Otting, Luck, Lawrence ve Burk yöntemleri ile SOD, CAT veselenyum bagımsız GSH-Px aktivitelerinin periyodik olarak tayini yapılmıstır.Elde ettigimiz bulgular sonucunda operasyon öncesinde tüm gruplarda enzimaktivitelerinin esit oldugu görülmüstür. Operasyon sonrasında kontrol grupları ilekarsılastırdıgımızda Atrisorb uygulanan bölgelerde en fazla olmak üzere, en az enzimaktiviteleri genelde lipozom ve atrisorb uygulanan bölgelerde görülmüstür. Grup içindedegerlendirme yaptıgımızda, tüm gruplarda, operasyon sonrası 3.günde enzim aktivitesiyükselerek pik seviyesine ulasmıs daha sonraki günlerde gittikçe azalmıstırSonuç olarak lipozom içine hapsedilmis SOD ve CAT kullanılarak PLA/PGAiçerikli biyomateryallerin biyoabsorbsiyonu/parçalanması sürecinde olusan reaktifoksijen türlerinin dokuda olusturabilecegi yan etkileri azaltmanın mümkün olabilecegiileri sürülmüstür.Anahtar Kelimeler : Serbest Oksijen Radikalleri, Serbest Radikal Süpürücü Enzimleri,Lipozomlar, Biyomateryaller, Diseti
The Activity Changes of SOD, CAT and Glutathione Peroxidase in AtrisorbApplied and Radiacal Scavenging Enzymes ?Superoxide Dismutase (SOD) andCatalase (CAT)? Delivered Periodontal Tissues of DogsABSTRACTAtrisorb is a polylactide acid(PLA)/polyglicolic acid (PGA) barrier that is used in aregenerative therapeutic method ?Guided Tissue Regeneration? to provide theregeneration of periodontal tissues lost by periodontitis.Former investigation reports have revealed reactive oxygen species (ROS)formation via ischema-reperfusion, inflammation and foreign substance applicationduring the wound healing period. Enduring these results, the study was aimed tominimise probable destructive effects of ROS on periodontal tissues by applyingliposom encapsulated SOD and CAT. The tissues were processed to assay SOD, CATand Glutathione Peroxidase (GSH-Px) activity changes in gingiva as an indicator ofROS for 1 month, periodically and the results analised to be informed about theformation of ROS in test and control groups, comparatively.The study performed on seven sistematically and periodontally healthy dogs (10-15kg) without the consideration of sexual discrimination. A commercially avaible soft dogfood was utilised for the nourishment of the animals and they were not treated with anykind of medicine during the study. The animals were kept at animal care complex andafter defect formation and stabilisation period, they were operated for regenerativetherapetic procedures by the inspection of a periodontolog. During preoperation andpostoperative therapy period, biopsies consisting of epitelial and connective tissue wereobtained from gingiva about 0.5 x 0.5 cm dimensions. The biopsies stored in 0.25Msucrose solution at -70 ºC in deep-freeze until the enzyme activity assay processesBefore the assay, biopsies processed with homogenisation, freze-thawing,sonication and santrifugation procedures to obtain supernatants. The supernatant wereutilised for the activity assays of SOD, CAT and selenium independent GSH-Px asshown by Mc Cord and Fridovich, Flohe and Otting, Luck, Lawrence and Burk,respectively.viiOur results revealed that activities of all enzymes were equal in all groups beforethe operation. After the operation activities were generally seen higher in atrisorbapplied groups and less in liposome and atrisorb applied groups, when compared withcontrol groups. Evaluating of all enzyme activities inside the groups were showed usthat after the operation activites were at peak point at 3. day and following days startedto decrease.In the light of these results it was concluded that SOD and CAT encapsulatedliposomes are useful to decrease damagive effects of ROS formed during thebiodegradation/bioabsorbsion of biomaterials consist of PLA/PGA.Key Words: Oxygen Free Radicals, Free Radical Scavenging Enzymes, Liposomes,Biomaterials, Gingiva.
