Abstract
ÖZET SAMSUN İLİNDE YETİŞTİRİLEN KABAKGÎL {CUCURBİTACEAE) BİTKİLERİNDE HASTALIK ETMENİ VİRÜSLERİN SAPTANMASI Samsun'da kabakgil üretiminin yapıldığı alanlarda virüs hastalıkları özellikle son yıllarda problem haline gelmiştir. Kabakgil bitkilerinde zararlı olan virüsleri ve bunların yayılışlarını belirlemek amacıyla, 1999 yılında Samsun' da 5 ilçeye bağlı 18 köyde 513 örnek toplanmıştır. Alınan örneklerin biyolojik ve serolojik yöntemler jle test edilmesi sonucunda bölgede Hıyar mozayik Cucumovirus (CMV)' unun, Kabak sarı mozayik Potyvirus (ZYMV)' unun ve Karpuz mozayik Potyvirus (WMV-2)' unun enfeksiyon oluşturduğu belirlenmiştir. DAS-ELISA yöntemi ile test edilen toplam 165 örneğin % 53.93' ünün WMV-2, %38.78' inin ZYMV ve %20.60' inin CMV ile bulaşık olduğu tespit edilmiştir. Ayrıca, CMV nin kapsid protein genine spesifik primerler ile enfekteli bitkilerde CMV enfeksiyonu reverse transkripsiyon-polimeraz zincir reaksiyonu (RT-PCR) yöntemi ile de belirlenmiştir. II ABSTRACT DETECTION OF VIRUSES INFECTING CUCURBITS [CUCURBİTACEAE) IN SAMSUN PROVINCE Viruses causing diseases in cucurbit crops has become problem in Samsun in recent years. To detect cucurbit viruses and find out their distrubition in the region, 513 samples were collected from the fields of eighteen villages in five districts in 1999. It was found that Cucumber mosaic Cucumovirus (CMV), Zucchini mosaic Potyvirus (ZYMV) and Watermelon mosaic Potyvirus-2 (WMV-2) were the most destructive viruses in cucurbits after analysing the samples by biological and serological methods. WMV-2, ZYMV and CMV were detected in 53.93%, 38.78% and %20.60 of 165 samples tested by DAS-ELISA, respectively. In addition to serological test, CMV was also determined in the infected plants by reverse transcription-polymerase chain reaction (RT-PCR) using primers specific to the capsid protein gene of CMV.
