Abstract
Bu araştırmada Samsun iline bağlı 15 ilçede yetiştirilen biber, domates, patates, patlıcan (Solanaceae), lahana (Brassicaceae) ve fiğ (Fabaceae) bitkilerinden elde edilen 70 Rhizoctonia izolatının geleneksel yöntemler ile gruplandırılması yapılmıştır. Elde edilen fungal izolatların Rhizoctonia solani AG 4, AG 7, Rhizoctonia zeae, BN Rhizoctonia AG-A, AG-F, AG-K, AG-S gruplarına ait oldukları tespit edilmiştir. Bu çalışmada elde edilen Rhizoctonia AG-A, AG-F, AG-K ve R. zeae izolatları fiğden elde edilmiştir. Ayrıca, Rhizoctonia AG-S izolatı patlıcandan, AG 7 izolatları ise patates bitkisinden elde edilmiştir. Ayrıca, hifal anastomoz reaksiyonlarına dayalı olarak gruplandırılan izolatları temsilen seçilen 20 izolatın ITS1-5.8S-ITS2 rDNA bölgesi dizi analizleri gerçekleştirilmiş ve izolatlar arasında ve alt gruplar içindeki genetik varyasyon belirlenmiştir. Üç farklı algoritma kullanılarak oluşturulan filogenetik analiz sonuçlarına göre R. solani AG 4 izolatları arasında HGI alt grubunun daha fazla çeşitlilik gösterdiği saptanmıştır. R. zeae izolatlarının kendi içinde iki alt dala ayrıldıkları tespit edilmiştir. BN Rhizoctonia AG-A izolatlarının ise iki soy hattına ayrıldığı belirlenmiştir. İzolatlar arasındaki genetik farklılığı belirlemek amacıyla ITS1-5.8S-ITS2 gen bölgesiyle yapılan değerlendirme sonucunda her anastomoz grubunu temsilen 9 izolat seçilmiştir Bu izolatların 18S rDNA bölgesine ait sekans verileri ve filogenetik analizlerine göre izolatların ayrımı tam olarak yapılamamıştır. In this study, 70 isolates of Rhizoctonia spp., which were obtained from pepper, tomato, potato, eggplant (Solanaceae), cabbage (Brassicaceae) and vetch (Fabaceae) plants grown in 15 districts of Samsun province, were grouped by the traditional methods. These fungal isolates were determined as belonging to the Rhizoctonia solani AG 4, AG 7, Rhizoctonia zeae, BN Rhizoctonia AG-A, AG-F, AG-K, AG-S according to anastomosis group reactions. Rhizoctonia AG-A, AG-F, AG-K and R.zeae isolates were recovered from vetch plant in this study. In addition, Rhizoctonia AG-S group and R. solani AG 7 isolates were isolated from eggplant and potato, respectively. Sequence of ITS1-5.8S-ITS2 rDNA regions were analyzed for the selected 20 isolates representing the isolates which were grouped on the basis of hyphal anastomosis reactions, and the genetic variations within sub-groups and among isolates were determined according to nucleotide sequence analyses. When considered variations in the trees of AG 4 isolates formed by using three different algorithms, sub-group HGI was found to be more variable than the other sub-groups. It was shown that both R. zeae and BN Rhizoctonia AG-A isolates were split into two sub-branches among themselves. In order to determine the genetic differences between isolates, nine isolates representing each anastomosis group were selected using ITS1-5.8S-ITS2 gene region data. From the trees generated using 18S rDNA sequence analysis, these isolates could not be differentiated exactly. Key words: Rhizoctonia spp., rDNA-ITS ,18S phylogeny, anastomosis groups.
