Bovine Herpesvirus Tıp 1 ve Herpes Simplexvirus-1 Üzerinde Porfirinin Antiviral Etkisinin Araştırılması

Abstract

Günümüzde kanserin fotodinamik terapisinde ve birçok hastalığın tedavisinde kullanılan porfirin ve türevleri bu çalışmada, Herpesviridae familyasında yer alan hem hayvanlar hem de insanlar için önemli enfeksiyonlara neden olan Bovine herpesvirus tip 1 ve Herpes simplexvirus-1 üzerinde antiviral etkileri açısından araştırılmıştır.Çalışma sırasında MDBK ve Vero hücre kültürlerinde porfirinin sitotoksik olmayan dozu hesaplanarak BHV-1 ve HSV-1 ile muamele edilmiş ve porfirinin apoptotik ve nekrotik hücreler açısından bu viruslar üzerindeki antiviral etkisi araştırılmıştır.Porfirin BHV-1 ile muamele edilip MDBK hücre kültürüne inokule edildiğinde, kontrol olarak kullanılan saf BHV-1'e karşı porfirinin virusu baskılayarak apoptoz ve nekrozu azalttığı tespit edilmiştir. Saf BHV-1 inokule edilen hücre kültüründe 6sa ve 24sa arası apoptoz %10 artış gösterirken porfirin+BHV-1 kombinasyonunda bu oran %5,3 olarak tespit edilmiştir. Nekroz açısından ise saf BHV-1'de 24sa sonunda nekrotik hücre sayısı %51 iken porfirin+BHV-1'de bu oranın %37,8'e düştüğü tespit edilmiştir.HSV-1'in Vero hücre kültürüne inokulasyonundan 24sa sonra apoptoz %23 iken porfirin+HSV-1 kombinasyonunda bu oran %22 olarak tespit edilmiştir. Nekrotik hücre değerlerine göre ise saf HSV-1, inokulasyondan 24sa sonra nekrozu %49 arttırmışken porfirin+HSV-1 kombinasyonunda bu oran %34 olarak bulunmuştur. Porphyrin and its derivates which are commonly used for the photodynamic treatment of cancer nowadays have been investigated for their antiviral effects on Bovine Herpesvirus Type-1 and Herpes Simplexvirus-1 which belong to Herpesviridae family and have importance for human and animal health.In this study, the non-cytotoxic dose of porphyrin was initially calculated, BHV-1 and HSV-1 were inoculated on MDBK and Vero cell lines respectively and the antiviral effect of porphyrin on these cell lines was researched.Comparing to Porphyrin treated BHV-1 to pure BHV-1 it is found that porphyrin has suppressed the virus and reduced apoptosis and necrosis of the cells. Between 6th and 24th hour of the inoculation of the pure BHV-1 the ratio of the apoptotic cells were raised 10% but 5.3% with the inoculation of porphyrin+BHV-1 combination. Furthermore, at 24th hour of the inoculation the necrotic cell ratio was 51% with pure BHV-1 while it was 37.8% with porphyrin+BHV-1 combination.On the other hand 24 hours after the inoculation of pure HSV-1 on Vero cell line the apoptotic cell ratio was 23% and necrotic cell ratio was 49% while it was 22% and 34% respectively with porphyrin+HSV-1 combination. According to these results porphyrin hasn?t been effective on the inhibition the HSV-1 in terms of apoptosis but it has made a change on the necrosis of the cells.Beyza YAVUZ, Master ThesisUniversity of Ondokuz Mayıs Samsun, May-2011
