Abstract

Bu çalışmada, ampisilin, ofloksasin, siprofloksasin, izoniazid, rifampisin, streptomisin ve etambutolün insan makrofajlarından nitrik oksit (NO) sentezine etkileri araştırılmıştır. Bu amaçla, Ficoll-Hypaque density gradient metod ile elde edilen ve 96 çukurlu plaklara dağıtılan insan makrofajları (104 makrofaj/ çukur) üzerine 1 \mug/ml ampisilin, 0.250 \mug/ml ofloksasin, 0.06 \mug/ml siprofloksasin, 1 \mug/ml izoniazid, 1 \mug/ml rifampisin, 2 \mug/m\ streptomisin ve 2 u\mug/ml etambutol eklenmiştir. Kültür süpernatanlarındaki nitrit seviyeleri 3., 6. ve 24. saatlerde Griess ayıracı ile ölçülmüş ve sonuç olarak izoniazid, ampisilin, ofloksasilin, streptomisin, etambutol içeren çukurlardaki nitrit seviyesinin önemli oranda arttığı gözlenmiştir.
The present study was carried out to determine the effects of ampicillin, ciprofloxacin, ofloxacin, isoniazid, rifampicin, streptomycin and ethambutol on the nitric oxide (NO) synthesis by human macrophages. Human macrophages were obtained by Ficoll-Hypaque density gradient method and after dispersing into the 96-well plate (104 macrophages per well), 1 \mug/ml ampicillin, 0.250 \mug/ml ofloxacin, 0.06 \mug/ml ciprofloxacin, 1 \mug/ml isoniazid, 1 u\mug/ml rifampicin, 2 \mug/ml streptomycin and 2 \mug/ml ethambutol were added to each well. The-nitrite levels in the culture supernatants were measured by Griess reagent at 3rd, 6th and 24th hours. As a result the nitrite levels were found to be significantly increased in the well containing isoniazid, ampicillin, ofloxacilin, streptomycin and ethambutol.
