Publication: Tavuk Genotiplerinde Genetik Uzaklığın RAPD (random Amplified Polymorphic DNA) Yöntemiyle Belirlenmesi
Abstract
TAVUK GENOTİPLERİNDE GENETİK UZAKLIĞIN RAPD (RANDOM AMPLIFIED POLYMORPHIC DNA) YÖNTEMİYLE BELİRLENMESİ ÖZET Ankara Tavukçuluk Araştırma Enstirüsü'nde yetiştirilen on saf hat; (RIR (Rhode Island Red) I, RIR II, Barred ( Barred Rock) I, Barred II, Colombian Rock (Col), Line-54, Blue Line (Blue), Maroon Line (Maroon), Black Line (Black), Brown Line (Brown), RAPD-PCR vasıtasıyla 12 universal primer kullanılarak araştırılmıştır. Hatlar arasında dokuz primer polimorfizm tespit etmiştir. Toplam olarak 35 bant tespit edilmiş ve sadece 15 bandın polimorfik olduğu bulunmuştur. Hatlar arasında en yüksek genetik yakınlık 0.3773 oranıyla Bar I ile Maroon hatları arasında, en az yakınlık ise 0.0896 oranıyla Col ile Bar II, Col ile RIR II, RIR II ile Maroon ve RIR II ile Black hatları arasında tespit edilmiştir. Dendogram, Nei'nin benzerlik katsayısı kullanılarak elde edilmiş, saf hatlar arasındaki benzer ilişkiler ortaya konulmuştur. Anahtar Kelimeler: Genetik uzaklık, RAPD, Tavuk
II DETERMINATION OF GENETIC DISTANCE BY RAPD (RANDOM AMPLIFIED POLYMORPHIC DNA) IN CHICKEN ABSTRACT Ten pure lines ((RIR (Rhode Island Red) I, RIR II, Barred ( Barred Rock) I, Barred II, Colombian Rock (Col.), Line-54, Blue Line (Blue), Maroon Line (Maroon), Black Line (Black), Brown Line (Brown)) breed in the Ankara Poultry Research Institute were investigated by RAPD-PCR using 12 universal primers. Only nine primers detected polymorphism between the lines.. To taly, 35 bands were scored and only 15 bands were found to be polymorphic. It is found that the highest genetic similarity value was at 0.3773 level between Bar I and Maroon; the lowest was at 0.8996 level between Col and Bar II, between Col and RTR II, between RIR II and Maroon, between RTR I and Black through the lines. Dendogram, derived using Nei's coefficient of similarity revealed similar relationships between the pure lines. Key Words: Chicken, Genetic distance, RAPD
