Nikotin ve Melatoninin Swiss Albino Tipi Laboratuvar Farelerinde Böbrek Antioksidant Enzim Aktiviteleri Üzerine Etkileri

Abstract

NİKOTİN VE MELATONİNİN SWISS ALBİNO TİPİ LABORATUVAR FARELERİNDE BÖBREK ANTİOKSİDANT ENZİM AKTİVİTELERİ ÜZERİNE ETKİLERİ ÖZET Bu çalışmada farelerde nikotin ve melatoninin böbrek antioksidan enzimlerinden olan süperoksit dismutaz (SOD) ve katalaz (CAT) aktiviteleri üzerine etkileri karşılaştırmalı olarak araştırılmıştır. Bu amaçla 24 saat aç bırakılan farelere nikotin ve melatonin intraperitonal olarak verildikten sonra 2., 4., 8., 12., 24. saatlerde servikal dislokasyonla öldürülerek böbrekleri çıkarılmıştır. Daha sonra bu böbreklere homojenizasyon, sonifıkasyon ve santrifugasyon işlemleri uygulanarak total enzim fraksiyonları elde edilmiştir. Bu fraksiyonlarda Lowry yöntemine göre protein tayini, SOD (Mc Cord ve Fridovich yöntemine göre) ve CAT (Flohe ve Otting ve Luck yöntemine göre) aktivite tayini yapılmıştır. Bu çalışmalar sonucunda, nikotinin etkisiyle böbrek CAT aktivitesinde genel bir inhibisyon görülmekle birlikte S.saatte çok küçük bir aktivasyon gözlenmiştir. Melatoninin etkisiyle böbrek CAT aktivitesinde sadece 2. saatte bir inhibisyon, diğer saatlerde ise genel bir aktivasyon gözlenmiştir. Nikotin ve melatoninin beraber etkisiyle böbrek CAT aktivitesinde 4. ve 8. saatte aktivasyon gözlenirken 2., 12. ve 24. saatte inhibisyon gözlenmiştir. Nikotinin etkisiyle böbrek SOD aktivitesinde 2.ve 4. saatte inhibisyon, 8., 12. ve 24. saatte ise aktivasyon gözlenmiştir. Melatoninin etkisiyle böbrek total SOD aktivitesinde tüm saatlerde aktivasyon gözlenmiştir. Melatonin ve nikotinin beraber verilmesiyle böbrek total SOD aktivitesinde 2., 4. ve 24. saatte inhibisyon, 8. ve 12. saatlerde ise aktivasyon gözlenmiştir. THE EFFECT OF NICOTINE AND MELATONIN ON KIDNEY ANTIOXIDANT ENZYME ACTIVITIES IN SWISS ALBINO TYPE LABORATORY MICE ABSTRACT In this study, the effect of nicotine and melatonin on the activites of superoxide dismutase and catalase as kidney antioxidant enzymes were researched comparatively in mice. For this purpose, before intraperitonal injection of nicotine and melatonin to labrotory mice kept hungry for 24 hours, and after the injection at the 2th,4th,8th, 12th and 24th hours mice were killed by servical dislocation, and their kidneys were removed. After total enzyme fractions were obtained by applying the kidneys homogenisation, sonification and centrifugation. These fractions were used for the determination of changes in superoxide dismutase (SOD) and catalase (CAT) activites. As a result, it was determined that the CAT activity generaly was inhibited but there was a few activation 8 th hour by the effect of nicotine. Under the effect of melatonin the CAT activity generally was activited but there was afew inhibition in 2th hour. And it was observed that under the effect of nicotine+melatonin the CAT activity activited 4th and 8th hours but inhibited 2th, 12th and 24th hours. it was determined that the SOD activity was inhibited 2th and 4th hours. But activited 8th, 12th and 24th hours under the effect of nicotine. Under the effect of melatonin, the SOD activity generally was activited all the hours. And it was observed that under the effect of nicotine+melatonin, the SOD activity inhibited 2th 4th and 24th hours but activited 8th and 12th hours.
