Abstract
Potao virus Y (PVY), biberde en fazla zarar oluşturan virüsler arasında yer almaktadır. Biber izolatları, pvr2- eIF4E lokusundaki pvr21 ve pvr22 dayanıklılık genlerine sahip biber genotiplerinin verdikleri reaksiyonlara göre üç ayrı patotip altında sınıflandırılmaktadır. Bu çalışmada, Samsun ilinde biberde enfeksiyon oluşturan PVY patotiplerinin belirlenmesi amacıyla 2010 ve 2011 yıllarında toplam 1012 yaprak örneği toplanmış ve daha sonra örnekler, PVY'ye spesifik poliklonal antiserum kullanılarak DAS-ELISA ile test edilmiştir. Test sonucunda 2010 yılında alınan örneklerin % 11.9'unun, 2011 yılında ise % 5.8'inin PVY ile enfekteli olduğu belirlenmiştir. İzolatların patotip ayrım setindeki bitkilerde oluşturduğu reaksiyonlara göre, % 80'nin PVY-0, % 20'sinin ise PVY-1 patotipi olduğu ortaya konulmuştur. PVY-0 ve PVY-1 patotiplerine ait ikişer izolatın CP gen bölgesinin baz ve aminoasit dizilişleri elde edilmiştir. Nükleotit BLAST analizi, PVY-0 patotipine ait SÇ-9 izolatının ve PVY-1 patotipine ait SÇ-84 ve SÇ-87 nolu izolatların, Tu12.3 biber izolatı ve LYE 84.2 domates izolatı ile benzerlik (% 97-98) gösterdiğini; PVY-0 patotipine ait SB-63 nolu izolatın ise patateste NTN ırkına ait bir çok izolat ile % 99 benzer olduğunu ortaya koymuştur. Ayrıca, 23 biber çeşidi ve 30 ıslah hattının PVY-0 ve PVY-1 patotiplerine karşı dayanıklılık düzeylerinin ve pvr2-eIF4E lokusundaki resesif pvr2 allel genlerinin belirlenmesine çalışılmıştır. Test edilen ıslah hatlarının tamamının her iki patotipe de hassas olduğu, çeşitlerden bir tanesinin PVY-0'a, 3 çeşidin ise her iki patotipe dayanıklı olduğu belirlenmiştir. Ayrıca, resesif pvr2 allellerine spesifik SNP (tek nükleotid polimorfizm) markörlerinin ve tetra primer amplification refractory mutation system (ARMS) PCR yönteminin dayanıklı çeşitlerin belirlenmesinde kullanılabileceği ortaya konulmuştur.
PVY is one of the most devastating viruses on pepper. Pepper isolates are classified into three groups according to the reactions of pepper genotypes possessing the pvr21 ve pvr22 resistance genes at the pvr2-elF4e locus. In this study, in order to determine PVY pathotypes infecting pepper in Samsun province, a total of 1012 leaf samples were collected in 2010 and 2011, and these samples were tested by DAS-ELISA using virus-specific polyclonal antiserum. The percentage of PVY infections were 11.9% in 2010 and % 5.8 in 2011. According to reactions of different pepper genotypes to these isolates after sap inoculation method, PVY pathotypes were determined, and among the isolates tested, PVY-0 pathotype (80%) was found to be more prevalent than PVY-1 (20%) in Samsun. The coat protein (CP) gene nucleotide and predicted aminoacid sequences of four isolates were obtained. Nucleotide BLAST analysis showed that SÇ-9 (PVY-0), SÇ-84 (PVY-1) and SÇ-87 (PVY-1) isolates of this study had sequence similaritiy (% 97-98) with Tu12.3 isolate from pepper and LYE 84.2 isolate from tomato, while the isolate SB-63 (PVY-0) had the highest similarity (% 99) with many isolates belonging to potato NTN strain. A total of 23 commercial pepper cultivars (cvs.) and 30 breeding lines were evaluated for the presence of pvr2 resistance alleles at the pvr2-eIF4E locus and resistance levels to PVY-0 and PVY-1 pathotypes. All breeding lines were determined to be sensitive to both pathotypes. Three commercial pepper cultivars were resistant to both PVY-0 and PVY-1, and however, one cv. was resistant to only PVY-0. Additionally, it was revealed that SNP (single nucleotide polymorphism) markers, which are spesific for the recessive pvr2 alleles, and the tetra primer amplification refractory mutation system (ARMS) PCR method were could be used to determine resistant varieties.
