Abstract
Bu araştırma, 2010-2012 yılları arasında Ondokuz Mayıs Üniversitesi Ziraat Fakültesi Bahçe Bitkileri Bölümünde, Amasya ve Giresun yörelerine ait 44 adet kiraz genotipinin S allellerini belirlemek amacıyla yapılmıştır. Bu genotiplerin S allellerinin belirlenmesinde, standart çeşitlerden S allelleri bilinen Starks Gold (S3S6), 0900 Ziraat (S3S12) ve MertonLate (S1S4) kontrol amaçlı olarak araştırmada kullanılmıştır. Araştırma' da uygulanan yöntem genel hatları ile kiraz çeşit ve genotiplerine ait yaprak örneklerinin DNA İzolasyonu, PCR işlemleri, Elektroforez ve elektroforez sonucu elde edilen bantların yorumlanması işlem basamaklarından oluşmaktadır. S allellerine özgü iki Forward, iki Reverse primer birbirleriyle kombine (Pru T2- Pru C4R, Pru C2- Pru C4R, Pru T2-SI32) edilerek kullanılmıştır. İncelenen genotiplerden 7 tanesinde kaliteli DNA elde edilememiş, 20 genotipte iki allel ve 15 adet genotipte ise tek allel belirlenmiştir.
This study was conducted in Ondokuz Mayıs University, Faculty of Agriculture Department of Horticulture between 2010 and 2012. The study aimed to determine the S alleles of 44 cherry genotypes in Amasya and Giresun regions. In the determination of S alleles of these genotypes, Starks Gold (S3S6), 0900 Ziraat (S3S12) and Merton Late (S1S4) whose S alleles are known were studied. The methods used in the study include DNA isolation of leaf samples belonging to cherry cultivars and genotypes, PCR processes, electrophoresis and interpretation of the bands obtained from electrophoresis. Two forward and two reverse primers specific to S-alleles were used in combination with each other (Pru T2- Pru C4R, Pru C2- Pru C4R, Pru T2-SI32). Of the genotypes investigated, good quality DNA could not be obtained in 7 genotypes; two alleles were determined in 20 whereas single allele was determined in 15 genotypes.
