Publication: Modifiye Atmosfer Paketli ve Hazırda Satılan Tavuk Döner Örneklerinde Salmonella Serotipleri ile İzolatlarda Antibiyotik Direnç Profilinin Belirlenmesi
Abstract
Amaç: Bu çalışmada, hazırda satılan ve modifiye atmosfer paketli dönerlerde Salmonella spp. varlığının klasik kültür tekniği kullanılarak araştırılması, elde edilen izolatların oriC gen bölgesi esas alınarak PCR ile doğrulanması ve izolatların serotiplerinin belirlenmesi ile antibiyotiklere karşı direnç profilinin saptanması amaçlanmıştır. Materyol ve Metot: Eylül-Kasım 2019 tarihleri arasında 3 ay süre ile toplam 150 tavuk döner örneği (75 hazırda satılan, 75 modifiye atmosfer paketli) analiz edildi. Örneklerdeki Salmonella spp.'ler klasik kültür tekniği ile izole edilerek şüpheli izolatlar MALDI-TOF yöntemi ile Salmonella spp. olarak belirlendi. PCR yöntemiyle izolatların konfirmasyonu tamamlanarak izolatların serotipleri belirlendi. İzolatların antibiyotik direnç profili disk difüzyon yöntemi ile saptandı. Bulgular: İncelenen 150 örnekten 23'ünün (%15,3) Salmonella spp. ile kontamine olduğu saptandı. Yapılan serotiplendirmede 33 izolatın tamamının S. Infantis olduğu belirlendi. Disk difüzyon testi sonucunda izolatların 30'u kloramfenikole, 24'ü gentamisine, 21'i ampisiline, 20'si trimetoprim-sulfametoksazol ve siproflaksasine, 18'i tetrasiklin ve amoksisilin-klavulanik asite, 13'ü ise sefoksitine karşı dirençli bulundu. Sonuç: Tavuk döner etlerinde Salmonella spp. izole edilmesi ve bu izolatların farklı antibiyotiklere direnç göstermesi halk sağlığı kapsamında önem arz etmektedir.

Salmonella spp. in the samples was isolated with classical culture. Suspicious isolates were determined as Salmonella spp. with MALDI-TOF method. Isolates were confirmed with PCR method. The resistance of the isolates against antibiotics were found with disc diffusion method. Results: Of the 150 samples analyzed, 23 (15,3%) were found to be contaminated with Salmonella spp. In the serotyping performed, all of 33 isolates were found to be S. Infantis. According to disc diffusion test results, 30 of the S. Infantis isolates was found to be resistant to chloramphenicol, 24 was found to be resistant to gentamicin, 21 was found to be resistant to ampicillin, 20 was found to be resistant to trimethoprim-sulfamethoxazole and ciproflaxacin, 18 was found to be resistant to tetracycline and amoxicillin-clavulanic acid and 13 was found to be resistant to cefoxitin. Conclusion: Isolation of Salmonella spp. in chicken doner meat and the resistance of the isolates to antibiotics is very important for public health.
