Beyaz Peynir Yüzeyinde Gelişen Mayaların İzolasyonu ve İdentifikasyonu

Abstract

iBEYAZ PEYN R YÜZEY NDE GEL ŞEN MAYALARIN ZOLASYONUVE DENT F KASYONUÖZETBu çalışmada beyaz peynir yüzeyinde gelişen ve peynirde potansiyel bozulmaetmeni olan maya suşlarının izole edilerek tanılarının yapılması amaçlanmıştır. Buamaçla, Samsun ilindeki farklı marketlerde satışa sunulan farklı marka veambalajlardaki peynirlerden 28 adet, evlerden sağlanan 12 adet olmak üzere toplam 40adet beyaz peynir örneği alındı. Peynir örnekleri Glikoz Pepton Yeast Extract Brothbesiyerine inoküle edilerek zenginleştirme işlemi yapıldıktan sonra petri kaplarına ekimyapılarak inkübasyona bırakıldı. nkübasyon sonunda gelişen kolonilerden rengi, şekli,büyüklüğü ve mikroskoptaki görünüşlerinin incelenmesi sonucunda 100 adet maya izoleedildi. Bu izolatlara biyokimyasal ve morfolojik tanı testleri uygulandı ve bu testlerdenelde edilen sonuçlar Lodder (1970), Kreger-Van Rij (1987), Barnett ve ark. (1990) veKurtzman ve Fell (2000)' in tanı anahtarı kullanılarak mayaların tanımlanması yapıldı.Tanımlama testlerinin sonucunda, izole edilen suşların % 22 Candida spp.,% 20 Pichia spp., % 16 Rhodotrula spp., % 14 Kluyveromyces spp., % 10 Cryptococcusspp., % 6 Metschnikowia spp., % 5 Debaryomyces spp., % 4 Yarrowia spp. ve% 3 Trichosporon spp. cinslerine ait türler olduğu düşünülmektedir.Bu sonuçlar, Candida spp., Pichia spp., Rhodotrula spp., Kluyveromyces spp. veCryptococcus spp.cinslerinin beyaz peynir yüzeyinden izole edilen dominant mayacinsleri olduğunu göstermektedir.Anahtar kelimeler: maya, peynir, izolasyon, tanımlama.

iISOLATION AND IDENTIFICATION OF YEASTS GROWN ON THESURFACE OF THE WHITE CHEESEABSTRACTIn this study, it is aimed at isolation and identification of yeast strains grown onthe white cheese surface and was an potential spoilage agent. For this purpose, total 40pieces samples of white cheese were collected. 28 samples belonging to different marksand packages were provided from different markets and the other 12 samples wereprovided from homes in Samsun. Cheese samples were inoculated into petri plates forenrichment procedure and then were left for the incubation. Total 100 yeast strains wereobtained in the result of the examination of color, shape, size and the microscopicappearances from growing colonies after incubation. Biochemical and morphologicidentification tests were applied for these isolates and yeasts were identified accordingto these tests results by using Lodder (1970), Kreger-Van Rij (1987), Barnett ve et.al.(1990) and Kurtzman and Fell (2000)? s identification key.At the results of identification test, yeast strains isolated are taught belonging tothe following genus. 22 % of them was Candida spp., 20 % Pichia spp., 16 %Rhodotrula spp., 14 % Kluyveromyces spp., 10 % Cryptococcus spp., 6 %Metschnikowia spp., 5 % Debaryomyces spp., 4% Yarrowia spp. and 3 % Trichosporonspp..These results showed that the genus of Candida spp., Pichia spp., Rhodotrulaspp., Kluyveromyces spp. and Cryptococcus spp.are those dominant yeast genusisolated on the surface of the white cheese.Key words: yeast, cheese, isolation, identification.
