Abstract
Bu çalışma kara turpun biyoaktif bileşenlerini ekstrakte etmek amacıyla yapılmıştır. Bu amaçla klasik ve ultrason destekli ekstraksiyon teknikleri kullanılmıştır. İki farklı teknikte çözücü, sıcaklık ve sürenin toplam fenolik madde miktarı ve antioksidan aktivite üzerine etkisi belirlenerek, ekstraksiyon çeşidine göre farklılıklar karşılaştırılmıştır. Belirlenen ekstraksiyon koşullarında en verimli uygulamanın klasik ekstraksiyonda çözücü: su, ultrasonik destekli ektraksiyonda (çözücü: su) 45°C ve 10 dakika ve ultrason destekli ekstraksiyonda (çözücü: etil alkol), 45°C ve 20 dakika olarak belirlenmiştir. Verimliliğin en yüksek olduğu noktalarda tahmin edilen toplam fenolik madde miktarı, FRAP ve EC50 değerleri sırasıyla klasik ekstraksiyon için 37.23 mg GAE/100 g, 2.14 mmol trolox/100 g ve 0.42 mg/mL, ultrason destekli ekstraksiyon (Çözücü: su) için 68.39 mg GAE/ 100 g, 2.46 mmol trolox/100 g ve 0.31 mg/mL ve ultrason destekli ekstraksiyon (Çözücü: Etil alkol) için 46.08 mg GAE/100 g, 3.36 mmol trolox/100 g ve 0.62 mg/mL 'dir. Ekstraktlarda en yüksek glukorafanin miktarı çözücü olarak su kullanılarak gerçekleştirilen ultrason destekli ekstraksiyon yöntemi ile elde edilmiştir. Kara turpun biyoaktif bileşenlerinin ekstraksiyonunda ultrason uygulamasının verimlilik açısından klasik uygulamaya kıyasla daha avantajlı olduğu belirlenmiştir.
The purpose of this study is the extraction of bioactıve components from the black radish. For this aim the classical extraction and ultrasound assisted extraction method were employed. In two different techniques, the effects of solvent type, extraction temperature, extraction time on total phenolic content and antioxidant activity of extract were detemined and results were compared. The most efficient method was water extraction method among the classical extraction methods; 45°C ve 10 min for ultrason - assisted extraction (using water as solvent) and 45°C ve 20 min for ultrason - assisted extraction (using ethanol as solvent).
