Abstract
Cyclosorine A (CsA) organ nakilleri ve otoimmün hastalıkların tedavisinde en yaygın kullanılan immün baskılayıcı ilaçtır. CsA'nın reaktif oksijen türleri üretimi ve lipit peroksidasyonu gibi hepatotoksik yan etkileri olduğu gösterilmiştir. Butylated hydroxyanisole (BHA), lipid peroksidasyonu zincirlerini kırabilme özelliğinden dolayı, gıdaların korunmasında en sık kullanılan fenolik bir antioksidandır.Bu çalışmada, CsA'nın neden olduğu oksidatif stresin hücresel yaşlanma üzerindeki etkilerinin belirlenmesi hedeflenmiştir. Ayrıca önemli bir antioksidan olan BHA vasıtasıyla CsA'nın hücresel yaşlanma üzerindeki etkilerinin indirgenmesi olasılığı da araştırılmıştır. Bu amaçla, her iki cinsiyet ve farklı yaş peryotlarından Wistar albino ratlar ile çalışma grupları oluşturuldu. 2 aylık (genç), 6 aylık (genç-ergin), 12 aylık (ergin) ve 24 aylık (yaşlı) erkek ve dişi ratlar dört gruba ayrıldı; Kontrol grubu (1), CsA uygulanmış grup (2), BHA uygulanmış grup (3) ve CsA+BHA uygulanmış grup (4). CsA (10 mg/kg/gün) intraperitonal olarak ve BHA (250 mg/kg/gün) gavaj yolu ile 7 gün boyunca uygulandı. Son uygulamadan 24 saat sonra her gruptan 4 erkek 4 dişi rat servikal dislokasyon işlemi ile öldürüldü. Karaciğer dokusundan alınan örneklerde, glutatyon ve malondialdehit miktarlarındaki; antioksidan enzimler süperoksit dismutaz (SOD), mitokondrial süperoksit dismutaz (Mn-SOD), katalaz (CAT) ve glutatyon peroksidaz (GPx) aktivitelerindeki değişimler spektrofotometrik olarak incelendi. Verilerin istatistiksel analizinde SPSS 15.0 programı kullanıldı.CsA özellikle yaşlı grupta GSH miktarında önemli bir azalmaya ve MDA miktarında, CAT, SOD ve Mn-SOD aktivitelerinde önemli bir artışa neden oldu. BHA, GSH miktarını arttırdı ve MDA miktarını azalttı. Birlikte uygulandıklarında BHA CsA'nın etkilerini azalttı.
Cyclosporine A (CsA) is the most widely used immunosuppressive drug in the organ transplants and in the treatment of autoimmune diseases. It has been shown that CsA has hepatotoxic side effects like as generating reactive oxygen species and lipid peroxidation. Cause of its ability to break the lipid peroxidation chains, Butylated hydroxyanisole (BHA) is frequently used fenolic antioxidant for protection of foods.In this study it has been aimed to determine effect of oxydative stress caused by cyclosporine A (CsA), on celluler aging. Additionally, it has also been investigated that the possibility of reducing the effects of CsA on celluler aging by using butylated hydroxianisole (BHA) which is an important antioxidant. For this aim sampling groups were created with Wistar albino rats which represents both gender and different age periods. 2 months old (young), 6 months old (young mature), 12 months old (mature) and 24 months old (old) male and female rats divided into four groups; Control group (1), CsA treated group (2), BHA treated group (3) and CsA+BHA treated group (4). CsA (10mg/kg/day) was treated by intraperitoneally and BHA (250mg/kg/day) was treated by gavage for 7 days. 24 hours after the last treatmet 4 male and 4 female rats from each groups were killed by cervical dislocation. In samples obtained from liver tissue, the amounts of Glutathione and malondialdehyde and changes in activities of antioxidant enzymes superoxide dismutase (SOD), mitochondrial superoxide dismutase (Mn-SOD), Catalase (CAT) and glutathione peroxidase (GPx) were determined by spectrophotometric. SPSS 15.0 programe were used for the statistical analysis of the data.CsA especially in the aged group caused a significant decrease in GSH level and significant increase in the activities of CAT, SOD, Mn-SOD and MDA level. BHA, increased the GSH and decreased the MDA levels. When they co-administered BHA decreased the effects of CsA.
