Publication: Siyatik Sinir Ligasyonu İle Oluşturulan Nöropatik Ağrı Modelinde Asiatik Asitin Etkisinin Erkek Sıçanlarda Araştırılması
Abstract
Nöropatik ağrı somatosensoriyel sistemi etkiyen bir hasar veya hastalıktan kaynaklanan ağrıdır.Nöropatik ağrı tedavisi zor, kullanılan ilaçların yan etkileri fazla, mevcut tedavilerin başarı oranı düşüktür ve yeni tedavi yaklaşımlarına ihtiyaç vardır. Çalışmamızda antiinflamatuvar, analjezik ve antioksidan etkisi bilinen bir triterpen olan asiatik asitin siyatik sinir ligasyonu ile oluşturulan nöropatik ağrıdaki etkisi araştırılmıştır. Siyatik sinir kontraksiyonu cerrahi yöntemle oluşturulduktan sonra 14 gün boyunca asiatik asit uygulanmış (3, 10 ve 30 mg/kg) sıçanlar 0., 7. ve 14. günde pençe ve hot plate testlerine tabi tutulmuştur. Mekanik allodini ve termal hiperaljezinin değerlendirilmesinden sonra deney hayvanları biyokimyasal ve moleküler olarak incelenmiştir. Ayrıca asiatik asitin nöropatik ağrı patolojisinde rol oynayan mikrogliozis ve nöron hasarı üzerindeki etkisi in vitro olarak BV2 ve SH-SY5Y hücre hatlarında araştırılmıştır. Asiatik asitin kronik tedavide ağrı yanıtını azalttığı ve spinal kord ve beyin düzeyinde antiinflamatuar etkiler ortaya çıkardığı görülmüştür. Ayrıca asiatik asitin siyatik sinir konstriksiyonu sonucu azalan BDNF seviyelerindeki azalmayı engelleyip normal seviyelere getirdiği, LPS ile indüklenen mikroglia hücreleri üzerinde toksik etkileri antiolsidan enzim seviyelerini arttırarak engellediği görülmüştür. Çalışmamızın sonuçları asiatik asitin siyatik sinir konstriksiyonu sonucu oluşan hasarı engelleyerek nöroprotektif etkisi olabileceğini göstermiştir.
Neuropathic pain is caused by damage or disease affecting the somatosensoryel nervous system. Neuropathic pain treatment is difficult, the drugs are high, the success rate of existing treatment approaches are needed in neuropathic pain. In our study, the effect of asiatic acid, a triterpene with known anti-inflammatory, analgesic and antioxidant effects, on neuropathic pain caused by sciatic nerve ligation was investigated. After the experimental model was created, the rats were treated with asiatic acid (3, 10 and 30 mg/kg) for 14 days and subjected to paw pressure and hot plate tests on days 0., 7, and 14. After the behavioral tests, the experimental animals were sacrificed with high dose anesthetic and biochemical moleculer analyzes were performed. In addition, the effect of asiatic acid on microgliosis and neurondamage, which plays a role in neuropathic pain pathology, was investigated in vitro in BV2 and SH-SY5Y cell lines. It has been observed that asiatic acid reduces pain response in chronic treatment and produces anti-inflammatory effects at the spinal cord and brain level. In addition, asiatic acid normalized the decreased BDNF levels as a result of sciatic nerve construction and prevented the toxic effects of LPS-induced microglia cells on neurons by increasing the levels of antioxidant enzymes. The results of our study show that asiatic acid may have a neuroprotective effect by protecting it from damage caused by sciatic nerve construction.
