Abstract
Bu çalıĢmanın amacı, seleksiyonla seçilmiĢ, çeĢide aday hatların ön verim denemeleri öncesi akrabalık iliĢkilerini belirleyerek hat sayısını azaltmaktır. Materyaller, daha önce bölgemizden topladığımız ve ulusal bitki gen bankasından elde ettiğimiz, morfolojik tanımlanması ve dokuz yıl süre ile seleksiyonu yapılmıĢ yerel bezelye gen kaynaklarıdır. ÇalıĢmada çeĢide aday 30 hat ve bu hatları kıyaslamak için 6 tane ticari çeĢit (Klein, Further, Lancet , Sprinter , G.Pearl, Vilmoren) kullanılmıĢtır. KıĢlık (15 Kasım) olarak ekilen bezelye genotiplerinin argonomik özelliklerinden bitki boyu, bitkide bakla sayısı, baklada tane sayısı, bitki ağırlığı, tane ağırlığı, çiçeklenme süresi ve kuru hasat süreleri tespit edilmiĢtir. Bu özelliklerin kullanılan genotiplerdeki değerleri sırarıyla 50.9-159.2 cm, 6.5-38.5 adet, 4.2-7.8 adet, 11.58-115.52 g, 41.26-224.32 g olmuĢtur. Bezelye genotipleri 27 SSR primer çifti ile moleküler olarak karakterize edilmiĢtir. AD186, AD126, AA67, AD141, A9, A5, AD148, AD83, AD61, AB141 ve AA122 SSR primerleriyle yapılan çalıĢmalardan değerlendirmeye uygun okumalar elde edilememiĢtir. Dolayısıyla AD270, AA278, B14, AD237, AA205, AA5, AA175, AC58, A6, AA90, AD56, AB23, AA285, AB72., AC76a ve AA446 numaralı SSR primer çiftlerinden elde edilen fragment analizi sonuçlarından yararlanılmıĢtır. UPGMA metoduna göre yapılan dendrogram ile akrabalık iliĢkisi oluĢturulmuĢtur. Elde edilen bu filogenetik ağaca göre hatlarımızdan Bz1-Bz2, Bz7-Bz8, Bz11-Bz12, Bz19-Bz20-Bz22-Bz23, Bz29-Bz30-G.Pearl, Bz37-Vilmoren arasında yüksek akrabalık belirlenmiĢtir. Ayrıca kontrol çeĢitlerden Lancet ile Sprinter arasında benzerlik yüksek bulunmuĢtur. Böylece 30 olan hat sayısı 24' e düĢmüĢtür. Bundan sonra ön verim denemelerine çıkılarak çeĢite aday hatlar tespit edilecektir.
This preliminary yield study was conducted with the aim of reducing number of pea varietal candidate lines by camparing similarity with previuos relative. material used in experiment was collected from local region, National Plant Gene Bank and from our own 9 years selection experiments on the bases of morphological identification were the main sources of seeds. In this research 30 candidate varietal lines and 6 commercial varieties (Klein, Further, Lancet, Sprinter, G.Pearl, Vilmoren) were used for comparison. Ġn winter season (15 November) pea genotypes were sown and agronomic parameters like plant height, number of pods per plant, number of seeds per pod, plant weight, seed weight, time taken to flowering and time taken to harvesting were recorded. These parameters were found 50.9-159.2 cm, 6.5-38.5 number, 11.58-115.52 g, 41.26-224.32 g, repectively. Pea genotypes 27 with SSR primer have been characterize as molecular pair. AD186, AD126, AA67, AD141, A9, A5, AD148, AD83, AD61, AB141 and AA122 lines assessment with SSR primer could not give appropriate readings. Therefore, AD270, AA278, B14, AD237, AA205, AA5, AA175, AC58, A6, AA90, AD56, AB23, AA285, AB72., AC76a ve AA446 number lines found get paired with SSR primer as a result of fragment analysis.. Dendrogram was created according to UPGMA method to represents the relationship of similirity.Resulting phylogenetic tree shows that strong relationship of similirity were found among Bz1-Bz2, Bz7-Bz8, Bz11-Bz12, Bz19-Bz20-Bz22-Bz23, Bz29-Bz30-G.Pearl and Bz37-Vilmoren lines. Furthermore, it was found very strong similarity between control varities Lancet and Sprinter. Therby 30 lines were reduced to 24.. After this study regional yield trial will be conducted using selected line as a candidate variety.
