Abstract
Bu çalışma sığır oositlerinin in vitro olgunlaştırılma (IVM) 'sında kültür medyumuna serum ilavesinin ısı şok protein 70 seviyesi üzerine etkisinin belirlenmesi amacıyla yapılmıştır. Çalışmada sığır kumulus oosit kompleksleri (KOK), fetal calf serum (% 10) ilave edilmiş ya da ilave edilmemiş bikarbonat tamponlu medyum-199'da 38,5 °C sıcaklıkta ve % 5 CO2'de nemlendirilmiş ortamda 22 saat IVM'na bırakılmıştır. IVM sonunda KOK'ların olgunlaşmaları tam kumulus genişlemesine göre belirlenmiştir. Olgunlaşan KOK'ların kumulus hücreleri % 0,1 hyarulinidaz enzimi içeren HEPES tamponlu TCM-199'da 10 dakika vortekslenerek uzaklaştırıldıktan sonra % 9,6'lık fosfat tamponlu tuz çözeltisinde (PBS) yıkanıp protein izolasyonuna geçilmiştir. Protein örneklerinin konsantrasyonunu arttırmak için örnekler 24 saat -80 °C'de dondurulduktan sonra liyofilizatörde 0.140 hPA basınç altında lyofilize edilip –20 °C'de depolanmıştır. Oositlerin 70 kDA ağırlıktaki proteinlerin seviyesi SDS- PAGE yöntemi ile belirlenmiştir. IVM'de serumlu (%84,78) veya serumsuz (%83,85) medyumlarda kültüre alınan oositler arasında kumulus genişlemesi bakımından bir farklılık tespit edilmemiştir. Western blot sonucunda kültür medyumuna serum ilavesiyle olgunlaştırılan sığır oositlerinin ısı şok 70 proteini bant görüntüsü elde edilemediğinden dolayı kültür ortamına eklenen serumun farklı sığır oositlerinin ısı şok proteinin 70 üzerine olabilecek etkileri tespit edilememiştir.
This study was conducted to determine the effect of serum addition to the culture medium on heat shock protein 70 production in in vitro maturation (IVM) of bovine oocytes. In the study, cattle cumulus oocyte complexes (COCs) were matured at bicarbonate buffered TCM-199 supplemented with and without 10% Fetal calf serum for 22 hours with a humidified 5% CO2 in air at 38.5 °C tempuratures. The maturation of COCs was determined according to the full cumulus expansion at the end of IVM. Cumulus cells of matured COCs were vortexed in HEPES buffered TCM-199 containing 0.1% hyarulinidase enzyme for 10 minutes and washed in 9.6% phosphate buffered saline (PBS) and protein isolation was started. To increase the concentration of the protein samples, the samples were frozen at -80 oC for 24 hours and lyophilized under lyophilizer at 0.140 hPA and stored at -20oC. The level of the proteins with a weight of 70 kD of oocytes was determined by SDS-PAGE. No difference was found between oocytes matured serum (%84,78) or serum-free (%83,85) cultured media in terms of fully cumulus cell expansion. As a result of the Western blot, heat shock 70 protein band images of bovine oocytes matured in serum or serum-free culture medium could not be determined.
