Abstract
2,2-Dikloropropionik asit (2,2 DCP), ksenobiyotik bir bileşik olup herbisit olarak kullanılmaktadır. Bakteriler herbisit ve insektisitlerin yoğun bir şekilde kullanıldığı Samsun'un Terme ilçesindeki bir tarım arazisinden izole edildi. İzole edilen izolatlar 2,2-dikloropropionik asiti yegâne karbon kaynağı olarak kullanmaktadır. Yapılan çeşitli biyokimyasal ve morfolojik testler sonucunda bu izolatların Pseudomonas' ın hangi suşuna ait olduğu belirlendi. Bununla birlikte genomik DNA izolasyonu yapılarak 16S rRNA metoduyla filogenetik ilişki açığa çıkarıldı. Bu sonuca göre bakteri %99 oranında Pseudomonas putida ve Pseudomonas mosselii bakterilerine benzerlik gösterildi. Bu bakteri dalaponlu minimal media besiyerinde yavaş bir şekilde büyüme gösterdi. Bu da bu bakterinin dalaponu karbon ve enerji kaynağı olarak kullandığının göstergesidir. Minimal media besiyerine 10 mM dalapon eklendi. Minimal media besiyerinden izole edilen bakterilerden genomik DNA izolasyonu yapıldı ve dehE geni PCR ile analiz edildi. İstenilen büyüklükte bantlar elde edildi.
2,2-dichloropropionate (2,2-DCP), is xenobiotic which used as herbicide. Bacteria were isolated from an agricultural area in the Terme district of Samsun where the herbicides and insecticides were used intensively. The bacteria was able to utilize 2,2 DCP as sole carbon source. As a result of various biochemical and morphological tests performed, it was determined which strain of these bacteria belong to Pseudomonas. Also, From16S rRNA analysis, the bacteria had 99% identity to Pseudomonas putida and Pseudomonas mosselii strains and shared many similarities in terms of biochemical reactions and microscopic observation. The bacteria was able to grow slowly in minimal medium contain only 2,2-dichloropropionate as sole carbon source and energy and growth were evident in minimal media containing 10mM 2,2 dichloropropionate. Genomic DNA isolation from bacteria isolated from minimal media medium was performed and dehE gene was analyzed by PCR. Bands of the desired size were obtained.
