Abstract
Ülkemizin tarım alanlarının %72'sinde tahıl üretimi yapılmaktadır. Diğer birçok tarımsal üründe olduğu gibi tahıllar içinde birçok fungal hastalık etmeni, ürün verimi ve kalitesini olumsuz yönde etkilemektedir. Bipolaris sorokiniana (Sacc.) Shoemaker (telemorf: Cochliobolus sativus), başak hastalığı (embriyo kararması), yaprak lekesi hastalığı (noktalı yaprak lekesi) vekök hastalığı (kök çürüklüğü) yapan buğday ve arpa üretimini kısıtlayan en önemli fungal hastalıkların başında gelmektedir. Bu çalışma ile ülkemizin birbirinden farklı agro-ekolojik bölgelerinde yetiştirilen arpa ve buğday bitkilerinde hastalık meydana getiren B.sorokiniana izolatlarının koloni rengi ve şekli, konidi boyutları ve bölme sayısı gibi özellikler dikkate alınarak morfolojik karakterizasyonu yapılmıştır. Aynı zamanda türe özgü primerler kullanılarak tür teşhisleride yapılmış ve bu türe ait olduğu belirlenen referans izolatlar seçilmiştir. Bu çalışmaların yanı sıra patojenite çalışmaları yürütülmüş, elde edilen sonuçlarla izolatların virülenslik düzeyleri ortaya konulmuştur. ITS bölgesi temeline dayalı kısmi gen sekanslaması yapılarak tür içindeki genetik farklılıklar belirlenmiştir. Eşleşme tipi (MAT) geni temelli eşey tiplemesi çalışması da yapılarak izolatların eşey tipleri belirlenmiştir. Grain production is being made in 72% of the agricultural area of our country. As in many other agricultural crops, many fungal diseases in grains affect negatively crop yield and quality. Bipolaris sorokiniana (Sacc.) Shoemaker (telemorf: Cochliobolus sativus) is one of the most important fungal diseases which causes ear disease, (black point), leaf disease (spot blotch) and root disease(common root rot) and restricts wheat and barley production. It has been determined with this study that the morphological characterization of B.sorokiniana isolates grown in different agro ecological regions of Turkey, taking into account the characteristics such as colony color and shape, conidia length and numbers of septa of B.sorokiniana isolates which cause serious diseases on barley and wheat plants. At the same time species identification of B.sorokiniana was carried out using species specific primers and reference isolates. In addition to these studies, pathogenicity tests were carried out and the results showed the virulence levels. Genetic differences within the species have been identified by partial gene sequencing based on the ITS region. Mating type (MAT) genes-based study was also conducted to determine the sexual types of isolates.
