Abstract
Bu çalışmada, en önemli endüstriyel ve çevresel kirleticilerden biri olan ve canlılar üzerinde çeşitli toksik etkilere neden olan kadmiyum ile güçlü bir antioksidan olarak bilinen selenyumun karaciğerde oluşturduğu histolojik değişiklikler ışık mikroskobu düzeyinde araştırılmıştır.Bu amaçla kadmiyum, selenyum, kadmiyum+selenyum ve kontrol grubu olmak üzere 4 grup hayvan kullanılmıştır. Ratlara (Wistar albino) kadmiyum ve selenyum gavaj yoluyla verildikten sonra 1., 6., ve 28. günlerde kardiyak perfüzyon işlemi uygulanarak karaciğerleri çıkarılmıştır.Karaciğerlerin fiksasyonu % 10'luk tamponlanmış nötral formalin ile yapıldıktan sonra rutin histolojik işlemler uygulanmıştır. Preparatlar hematoksilen-eosin (H-E), Masson'un üçlü boyaması ve periodik asit-schiff (PAS) teknikleri ile boyanmıştır.Çalışmamızdan elde edilen sonuçlara göre; kadmiyumun etkisiyle, karaciğerde hidropik dejenerasyon, sinüzoidlerde genişleme, inflamasyon odakları gözlenmiştir. Sadece selenyum verilen gruplarda sinüzoidlerde hafif şiddette genişlemeler dışında karaciğerde belirgin bir değişiklik olmamıştır. Kadmiyum ve selenyumun birlikte verildiği gruplarda ise, sadece kadmiyum verilen gruplara göre daha az histolojik değişiklikler gözlenmiştir.Bu bulgular, bir serbest radikal süpürücüsü olan selenyumun, kadmiyumun serbest radikal oluşumunu indükleyerek karaciğerde meydana getirdiği oksidatif hasarı azalttığını göstermektedir.
In this study, histologic changes induced by cadmium which is the most important industrial and environmental pollutive and caused various toxic effects in life and selenium which is known as a strong antioxidant, were investigated under light microscopy. For this purpose, four groups of animals were used as selenium, cadmium, cadmium+selenium and control groups. Cadmium and selenium were given via gavage rats (Wistar albino) , they were killed at 1th, 6th, 28th days after cardiac perfusion and their livers were removed. After isolated livers were fixed in % 10 buffered neutral formalin, routin histological processes were applied and sections were stained by hematoxilen-eosin (H-E), Masson?s trichrome and Periodic asit-Schiff (PAS).According to results obtained in our study; histologic changes such as hydropic degeneration, dilation at sinuzoids and inflammatory focuses induced by cadmium were determined. It was seen that using only selenium didn?t cause any obvious differentiation on liver except mild sinüzoidal dilation. When cadmium and selenium were given together, the degree of histological changes induced by cadmium and selenium, was less than cadmium alone.These findings show that selenium, as a free radical sweeper, decreases the oxidative damage that cadmium causes in liver by induced free radical formation.
