Abstract
ASPİRİN VE VİTAMİN E (a-Tokoferol)'NİN FARELERDE {Mus musculus) KARACİĞER TOTAL SÜPEROKSİT DİSMUTAZ VE KATALAZ AKTİVİTELERİNE ETKİLERİ ÖZET Bu çalışmada Aspirin ve en önemli antioksidant olan E vitamininin, radikal süpürücü enzimlerden Katalaz (CAT) ve Süperoksid dismutaz'ın (SOD) aktivitesi üzerindeki etkileri araştırılmıştır. Bu amaçla 24 saat aç bırakılan laboratuvar farelerine (Mus musculus) Aspirin ve E vitamini intraperitonal yolla enjekte edildikten sonra 2.,4.,8.,12.,24 ve 48. saatlerde servikal dislokasyon yoluyla öldürülerek karaciğerleri çıkarılmıştır. Daha sonra bu karaciğerlere sırasıyla homojenizasyon, sonifikasyon ve santrifügasyon işlemleri uygulanarak, total enzim fraksiyonu elde edilmiştir. Bu fraksiyonda Lowry yöntemine göre protein miktarı belirlendikten sonra Katalaz ile Süperoksit dismutaz aktiviteleri ölçülmüştür. Bu çalışmalar sonucunda, Aspirin ve Vitamin E'nin ayrı ayrı ve birlikte etkileri sonucu Karaciğer Süperoksit dismutaz (SOD) aktivitesinin inhibe edildiği belirlenmiştir. Aspirin ve Vitamin E'nin birlikte etkileri ile karaciğer Katalaz aktivitesi inhibe edilirken Vitamin E'nin aktivasyona neden olduğu belirlenmiştir. THE EFFECTS OF ASPIRIN AND VITAMIN E (a-Tocopherol) ON THE TOTAL ACTIVITIES OF LIVER SUPEROXID DISMUTASE AND CATALASE IN MICE (Mus musculus) ABSTRACT In this study, effects of Aspirin and Vitamin E which is the most important antioxidant on the radical-scavenger enzymes Catalase and Superoxide dismutase activities were investigated. For this purpose, after interperitonal injection of Aspirin and Vitamin E to laboratory mice kept hungry for 24 hours, this animals were killed by servical dislocation at 2, 4, 8, 12, 24 and 48th hours and their livers were isolated. Then, these livers were exposed to homogenisation.sonification and centrifugation. Thus total enzyme fraction were yielded. In this fraction, after determining protein amount by Lowry method, Catalase and Superoxide dismutase activities were measured. In this study, it was coucluded that the Superoxide dismutase activity was inhibited by Aspirin and Vitamin E together and individually. While the liver Catalase activity was inhibited by the effect of Aspirin and Vitamin E, it was determined that the Catalase activity was activated by the effect of Vitamin E individually.
