Abstract
ÖZET DACTYLORHIZA URVILLEANA (STEUDEL) BAUMAN VE KÜNKELE VE D. IBERICA ( BIEB.EX WILLD ) SOÖ ( ORCHIDACEAE ) TÜRLERİNİN KÖKLERİNDEN FUNGUSLARIN İZOLE EDİLMESİ, BU TÜRLERE AİT TOHUMLARIN SİMBİYOTİK VE ASİMBİYOTİK KÜLTÜR ORTAMLARINDA ÇİMLENME VE GELİŞMESİ ÜZERİNDE BİR ARAŞTIRMA Bu çalışmada Dactylorhlza urvllleana ve D. Iberica'mn tohumlarının farklı sodyum hipoklorit solüsyonlarında yüzeysel sterilizasyonu yapılmıştır. En iyi çimlenme ve gelişme için yüzeysel sterilizasyon solüsyonunun konsantrasyonu ve uygulama zamanının, orkide türlerine göre değiştiği belirlenmiştir. Simbiyotik kültür çalışmalarında, iki türün köklerinden izole edilen fungal izolatlar ve yurtdışından sağlanan izolatlar kullanılmış, fungal izol ati arın etkililik derecelerinin kültür ortamının besinsel içeriğine göre değişebildiği belirlenmiştir. Asimbiyotik çimlendirme çalışmalarında seyreltik ve konsantre kültür ortamları kullanılmıştır. Seyreltik kültür ortamlarına eklenen şekerin çimlenme üzerinde olumsuz etki yaptığı belirlenmiştir. Konsantre Van Waes-Deberg kültür ortamı ve bu ortamın modifiye edilmesiyle hazırlanan ortamlarda, şeker ve inorganik azotun daha iyi fide gelişimi için gerekli olduğu belirlenmiştir. Yüksek oranda çimlenme elde edilen kültür ortamlarındaki protokormların birkaçı, daha sağlıklı fide gelişiminin ve köklü fidelerin meydana geldiği ortamlara transfer edilmiştir.
ABSTRACT AN INVESTIGATION ON THE ISOLATION OF THE FUNGI FROM ROOTS OF DACTYLORHIZA URVILLEANA (STEUDEL) BAUMAN END KÜNKELE AND D. IBERICA (BIEB.EX WILLD) SOÖ (ORCHIDACEAE), THE GERMINATION AND DEVELOPMENT OF THE SEEDS OF THIS SPECIES IN THE SYMBIOTIC AND ASYMBIOTIC CULTURE MEDIA. In this study, the seeds of Dactylorhiza urvilleana and D. iberica were treated with different concentration of sodiumhypochlorid for surface sterilization. It was found that for the best germination and seedling development concentration of the sterilization solution and the application period were not the same for different orchid species. The effectiveness of the fungal isolates which were isolated from the roots of the two species mentioned above and the fungal isolates which were obtained from the other countries was not the same with different nutritional components in the symbiotic media. In the asymbiotic germination experiments diluted and concentrated media were used. It was determined that effect of sugar was negative on the germination in the diluted media. On the other hand in the concentrated Van Waes-Deberg culture medium and in the modified form of this, sugar and inorganic nitrogen was necessary for better seedling development. In the asymbiotic culture media protocorms were transferred to the other media in which protocorms reached to a better seedling development of the rooted seedling stage.
