Abstract
Kirazın en önemli bakteriyel hastalıkları arasında bakteriyel kanser hastalık etmenleri Pseudomonas syringae pv. syringae ve Pseudomonas syringae pv. morsprunorum gelmektedir. Bu etmenler kirazın çiçek, meyve, yaprak, dal ve gövdesinde belirti oluşturmaktadır. Özellikle dal ve gövdede meydana getirdikleri kanserler nedeniyle ağacın ölümüne yol açmaktadırlar. Bu çalışma 2016-2017 yılları arasında Amasya İli kiraz yetiştiriciliği yapılan bahçelerde kiraz bakteriyel hastalık etmenlerini belirlemek amacıyla yapılmıştır. Survey çalışmaları ile kirazın çiçek, yaprak ve kabuk dokularından yapılan izolasyonlar sonucunda toplam 103 adet izolat elde edilmiştir. Bu izolatlardan 5 adeti tütünde aşırı duyarlılık reaksiyonuna, 23 adeti ise hem tütünde aşırı duyarlılık reaksiyonuna hem de kirazda patojenisite testine pozitif reaksiyon göstermiştir. Kirazda patojenisite testine negatif reaksiyon gösteren 5 adet izolatımn morfolojik, biyokiyasal ve moleküler tanısı sonucunda: Pseudomonas graminis, Bacillus simplex, Pantoea agglomerans, Pseudomonas poae ve Curtbacterium flaccumfaciens türlerine ait epifitik izolatlar olduğu: hem tütünde aşırı duyarlılık testine hem de kirazda patojenisite testine pozitif reaksiyon gösteren 23 adet izolatın ise morfolojik, biyokiyasal ve moleküler tanısı sonucunda Pseudomonas syringae pv. syringae olduğu belirlenmiştir.
Bacterial canker agents, Pseudomonas syringae pv. syringae and Pseudomonas syringae pv. morsprunorum are including cherry's most important bacterial disease agents. Disease symptoms of these agents on the cherry flowers, fruits, leaves, branch and trunk. Specially, the bacterial canker agents cause dieback and canker on branches and trunks. This study was carried out between 2016 and 2017 in Amasya. The aim of the study is to determine the cherry bacterial disease agents in cherry gardens. As a result the surveys, 103 isolates were obtained. The isolates were obtained from the flowers, leaves and bark of the branch and trunk.Five of these isolates had a positive reaction to the hypersensitive reaction and 23 of them had a positive response to both the hypersensitive reaction tobacco and the pathogenecity test on cherry leaves. Morphological, biochemicial and molecular identification of 5 epiphytic isolates showing a negative reaction as a pathogenicty test on the cherry leaves were performed. The names of these isolates were identified as Pseudomonas graminis, Bacillus simplex, Pantoea agglomerans, Pseudomonas poae and Curtobacteriım flaccumfaciens. 23 isolates had positive responsse to hypersensitive reaction on tobacco and pathogenicity test on cherry leaves. These isolates were identified according tomorphological, biochemicial and molecular tests as Pseudomonas syringae pv syringae.
