Abstract
SİSPLATİNİN RAT (Wistar albino) KARACİĞERİNDE OLUŞTURDUĞU HİSTOLOJİK HASAR ÜZERİNE ANTİOKSİDAN BETA-GLUKANIN ETKİLERİNİN IŞIK MİKROSKOBU DÜZEYİNDE ARAŞTIRILMASIÖZETBu çalışmada, antineoplastik bir ajan olan sisplatin ile güçlü bir antioksidan ve immünomodülatör olduğu bilinen beta-glukanın karaciğerde oluşturduğu histolojik değişiklikler araştırılmıştır.Bu amaçla sisplatin, beta-glukan, sisplatin+beta-glukan ve kontrol grubu olmak üzere 4 grup hayvan kullanılmıştır. Ratlara (Wistar albino) sisplatin ve beta-glukan intraperitonal enjeksiyonla verildikten sonra 1., 2., 7., ve 14. günlerde servikal dislokasyonla öldürülerek karaciğerleri çıkarılmıştır.Karaciğerlerin fiksasyonu %10' luk tamponlanmış nötral formalin ile yapıldıktan sonra rutin histolojik işlemler uygulanmıştır. Preparatlar hematoksilen-eosin (H-E), Masson'un üçlü boyaması ve periodik asit-schiff (PAS) teknikleri ile boyanmıştır.Çalışmamızdan elde edilen sonuçlara göre; sisplatinin etkisiyle, karaciğerde hidropik dejenerasyon, sinüzoidlerde genişleme, inflamasyon odakları tespit edilmiştir. Sadece beta-glukan verilen grupta karaciğerde belirgin bir değişik olmamıştır. Sisplatin ve beta-glukanın birlikte verildiği gruplarda, sadece sisplatin verilen gruplara göre daha az histolojik değişiklik gözlenmiştir. Ayrıca, sisplatin+beta-glukan 14. gün gruplarındaki histolojik görünümün, sisplatin+beta-glukan 7. gün gruplarındaki ile karşılaştırıldığında normale daha yakın olduğu görülmüştür.Bu bulgular, bir serbest radikal süpürücüsü olan beta-glukanın, sisplatinin serbest radikal oluşumunu indükleyerek karaciğerde meydana getirdiği oksidatif hasarı azalttığını göstermektedir.Anahtar kelimeler: Sisplatin, beta-glukan, karaciğer, histolojik hasar.
LIGHT MICROSCOPIC INVESTIGATION OF ANTIOXIDANT BETA-GLUCAN?S EFFECTS ON CISPLATIN INDUCED LIVER HISTOLOGIC DAMAGE IN RAT (Wistar albino)ABSTRACTIn this study, histologic injury induced by cisplatin which is an anti-neoplastic agent and ß-glucan which is known as a strong antioxidant and immunomodulator, were investigated. For this purpose, four groups of animals were used as ß-glucan, ciplatin, Cisplatin + ß-glucan and control groups. Cisplatin and ß-glucan were injected rats (Wistar albino) via intraperitonally, they were killed after at 1th, 2th, 7th, 14th days by servical dislocation and their livers were removed. After isolated livers were fixed in % 10 buffered neutral formalin, routin histological processes were applied and sections were stained by hematoxilen-eosin (H-E), Masson?s trichrome and Periodic asit-Schiff (PAS).According to results obtained in our study; histologic changes such as granular degeneration, dilation at sinuzoids and inflammatory focuses induced by cisplatin were determined. It was seen that using only ß-glucan didn?t cause any obvious differentiation on liver. When cisplatin and ß-glucan were given together, the degree of histological changes induced by cisplatin and ß-glucan, was less than cisplatin alone. It was seen that the histological picture was more close to normal in the cisplatin + ß-glucan 14th day groups when compared with cisplatin + ß-glucan 7th day groups.These findings show that ß-glucan, as a free radical sweeper, decreases the oxidative damage that cisplatin causes in liver by induced free radical formation.Key Words: Cisplatin, beta-glucan, liver, histological injury.
