Abstract
Bu çalışmada, Kuzey Kıbrıs Türk Cumhuriyeti?nin 3 farklı lokalitesinden Streptomyces cinsi üyesibakteri izolasyonu yapılmıştır. İzolatların 16S rRNA, atpD, gyrB, recA, rpoB ve trpB gen bölgelerine ait nükleotit dizileri elde edilmiş ve filogenetik sistematikleri yapılmıştır. İzole edilen toplam 40 izolatın renk gruplandırması yapılarak 9 renk grubu oluşturulmuş ve her renk grubundan temsilci bir izolat seçilerek bu izolatların 16S rRNA gen bölgesinin PCR çalışmaları gerçekleştirilmiştir. 16S rRNA gen bölgesi nükleotit dizi analizi sonuçlarına göre izolatların Streptomyces cinsi üyesi oldukları belirlenmiştir. 16S rRNA gen bölgesi nükleotit dizi analizi ile oluşturulan filogenetik dendogramda izolatların en yakın komşu tip türleri tespit edilmiştir. Karşılaştırmalı multi gen dizi analizi çalışmalarının yapılması için tip türlerinin atpD, gyrB, recA, rpoB ve trpB gen bölgeleri nükleotit dizileri NCBI gibi veri bankalarından elde edilmiştir. Gen bölgesi sekans verileri bulunmayan tip türleri çeşitli (DSMZ, KCTC gibi) kültür koleksiyonlarından elde edilerek beş gen bölgesi nükleotit dizi analizleri gerçekleştirilmiştir. Multi gen dizi analizi (MLSA=Multilocus Sequence Analysis) çalışmasında kullanılan gen bölgelerinin filogenetik analizleri sonucunda test izolatlarının tip türlerinden nükleotit farklılıkları ve filogenetik uzaklıkları belirlenmiştir. Ayrıca 73 birim karakter bakımından test izolatları ile tip türleri nümerik sonuçlarla karşılaştırılmıştır. Yapılan çalışmalarda farklı iki tür arasında MLSA filogenetik uzaklık değerinin 0,007?den yüksek olması gerektiği belirtilmiştir. 9 test izolatından 7 tanesinin tip türlerinden MLSA filogenetik uzaklık değerinin 0,007?den yüksek olduğu tespit edilmiştir. Ayrıca fenotipik farklılıklar ve gen bölgeleri nükleotit dizilerine dayalı oluşturulan dendogramlardaki yüksek bootstrap değerleri bunu desteklemektedir.Sonuç olarak; K217, K330, K326, K112, K108, K324 ve K101 izolatlarının yeni tür oldukları tahmin edilmektedir. In this research, bacteria that members of genus Streptomyces, isolated from three different localities of Turkish Republic of Northern Cyprus. Nucleotide sequences belong to 16S rRNA, atpD, gyrB, recA, rpoB and trpB regions obtained and was made phylogenetic systematics of isolates. Color grouping were made in these 40 isolates and nine color groups were formed, and by selecting a represantative isolate from each color groups, PCR studies of 16S rRNA gene of these isolates were performed. According to 16S rRNA gene region nucleotit sequence analysis, isolates were determined to be a members of genus Streptomyces. In phylogenetic dendogram, generated by 16S rRNA gene region nucleotide sequence analysis, the nearest neighbour type species of these isolates were determined. In order to make the comparative multi locus sequence analysis studies, atpD, gyrB, recA, rpoB and trpBgene regions nucleotide sequences were obtained from data banks such as NCBI. Type species which have not gene region sequence datas, were obtained from various (such as DSMZ, KCTC) culture collections and five gene regions nucleotit sequence analysis were performed. In the results of phylogenetic analysis of gene regions used in the study of multi locus sequence analysis (MLSA), it can be determined evolutionary distances and nucleotit differences of test isolates from type species. Furthermore, test isolates and type species were compared with numerical results with regard to 73 unit character. It has been reported in the literature that MLSA evolutionary distance value should be higher than 0,007 between different two species. 7 of 9 test isolates was higher than 0,007 MLSA evolutionary distance value from type species. Besides, it was supported with phenotypic differences and high bootstrap values in dendograms formed by gene regions nucleotit sequences. As a result of this research, it is estimated that K217, K330, K326, K112, K108, K324 and K101 isolates are new species. Key Words: Streptomyces, 16S rRNA, MLSA, Phylogenetic Systematics.
