Abstract
Akrolein, insanların özellikle duman gibi çeşitli çevresel durumlarda maruz kaldığı son derece reaktif ?,ß-doymamış aldehittir. Beta-Glukan (ß-Glukan), maya, mantar, bakteri ve tahıl bitkilerinin hücre duvarında bulunan glukoz polimerleridir. İmmün sistem üzerindeki faydalı etkilerinden dolayı ß-glukan üzerine çeşitli çalışmalar uygulanmaktadır.Bu çalışmada, akrolein ve ß-glukanın rat karaciğerinde glutatyon (GSH) ve malondialdehit (MDA) düzeyleri, süperoksit dismutaz (SOD), katalaz (CAT), glutatyon peroksidaz (GPx) ve kaspaz 3 aktiviteleri üzerine etkilerini araştırdık. Çalışmada ergin dişi ve erkek Wistar albino ratlar kullanıldı. Ratlar, 4 gruba bölündü: Grup I (kontrol) , Grup II (ß-glukan), Grup III (akrolein) ve Grup IV (akrolein + ß-glukan). ß-glukan (50 mg/kg/gün) ve akrolein (4 mg/kg/gün), 5 gün süreyle tek dozda gavajla verildi. Her gruptan 5 dişi ve 5 erkek rat, son gavajdan sonra 6., 12., 24. ve 48. saatlerde servikal dislokasyon ile öldürüldü. Biyokimyasal analizler için karaciğer doku örnekleri alındı. Doku örneklerinde GSH ve MDA düzeyleri, SOD, CAT, GPx ve kaspaz 3 aktiviteleri spektrofotometrik yöntemlerle belirlendi. Veriler, SPSS 15.0 ile istatistiksel olarak değerlendirildi.Akrolein ve ß-glukan grupları kontrol grubu ile karşılaştırıldığında zamana bağlı olarak SOD, CAT ve GPx enzim aktivitelerinde artışa, GSH seviyesinde önemli artış ve azalmaya neden oldu. Akrolein MDA düzeyini artırırken, ß-glukan azalmaya neden oldu. Ayrı ayrı uygulandıklarında kaspaz 3 aktivitesinde herhangi bir değişikliğe neden olmazken, birlikte uygulandıklarında artışa neden oldular.Anahtar Kelimeler: Akrolein, ß-glukan, GSH, MDA, SOD, CAT, GPx, Kaspaz 3
Acrolein is a highly electrophilic ?,ß-unsaturated aldehyde to which humans are exposed in a variety of environmental situations, particularly as smoke. ß-glucans are glucose polymers found in the cell walls of yeast, fungi, bacteria and cereal plants. A variety studies were performed on ß-glucan molecule because of the beneficial effects on the immune system.In this study, we investigated the effects of acrolein and ß-glucan on GSH and MDA levels, SOD, CAT, GPx and caspase 3 enzyme activities in rat liver. In the study, adult female and male Wistar albino rats were used. The rats were divided into 4 groups: Group I (Control), Group II (ß-Glucan), Group III (Acrolein) and Group IV (Acrolein + ß-Glucan). ß-glucan (50 mg/kg/day) and acrolein (4 mg/kg/day) were given by gavage a dose for 5 days. 5 females and 5 males rats in each group were killed by cervical dislocation at 6th, 12th, 24th and 48th hours after last gavage. The tissue samples of liver were obtained for biochemical analyses. In tissue samples, GSH and MDA levels, SOD, CAT, GPx and caspase 3 activities were determined by spectrophotometric methods. The data were evaluated statistically using SPSS 15.0.Acrolein and ß-glucan groups caused as a time-dependent a significant decrease and increase in the GSH level, increase in the SOD, CAT and GPx activities when compared with the control group. While acrolein elevated the MDA level, ß-glucan induced to decreased. They did not cause any change in caspase 3 activity administered separately.
