Abstract
Bu çalışma 2008-2009 yıllarında Samsun ilinin Bafra, Vezirköprü, Alaçam, Tekkeköy ilçelerinde tütün alanlarında enfeksiyon oluşturan virüslerin saptanması ve bulaşıklık oranlarının ortaya konulması amacıyla yürütülmüştür. Bu ilçelere bağlı 15 köyde, toplam 28 tarladan virüsle enfekteli olduğundan şüphe edilen toplam 196 adet tütün örneği toplanmış ve bu örnekler double antibody sandwich-enzyme linked immunosorbent assay (DAS-ELISA) yöntemi ile test edilmiştir.Test sonucunda, örneklerin büyük bir çoğunluğunda (% 40.3) Tobacco mosaic virus (TMV), enfeksiyonu ile karşılaşılmıştır. Bunu sırasıyla % 7.65 ile Potato virus Y (PVY), % 5.61 ile Tomato spotted wilt virus (TSWV), % 4.08 ile Tobacco etch virus (TEV) enfeksiyonları izlemiştir. Çok az sayıda bitki (% 2.04) Cucumber mosaic virus (CMV) veya Alfalfa mosaic virus (AMV) ile bulaşık olarak belirlenmiştir. Tobacco necrosis virus (TNV) ve Tobacco ring spot virus (TRSV) ile bulaşık hiçbir örneğe rastlanmamıştır. Çok az oranda da olsa bazı bitkilerin ikili veya üçlü viral enfeksiyonlara sahip olduğu saptanmıştır.Anahtar kelimeler: ELISA, virus, tütün.
This study was conducted during 2008-2009 to detect the presence of viral pathogens and determine their incidence in tobacco fields in Bafra, Vezirköprü, Alaçam and Tekkeköy districts of Samsun Province. A total of 196 samples were collected from 28 fields in 15 villages and tested by double antibody sandwich-enzyme linked immunosorbent assay (DAS-ELISA).The results showed that most of the samples (40.3%) were infected with Tobacco mosaic virus (TMV). The percentage of samples infected with Potato virus Y (PVY), Tomato spotted wilt virus (TSWV) and Tobacco etch virus (TEV) were 7.65%, 5.61% and 4.08%, respectively. Only 2.04% of the samples were infected with either Cucumber mosaic virus (CMV) or Alfalfa mosaic virus (AMV), but none of the samples were found to be positive for Tobacco necrosis virus (TNV) and Tobacco ring spot virus (TRSV).
