Abstract
BETA-GLUKAN VE TAKSOL ETK S YLE FARE (Mus musculus)KARAC ĞER NDE OLUŞAN H STOLOJ K DEĞ Ş KL KLER N IŞIKM KROSKOBU DÜZEY NDE ARAŞTIRILMASIÖZETBu çalışmada, antineoplastik bir ajan olan taksol ile güçlü bir antioksidanve immünomodülatör olduğu bilinen beta-D-glukanın karaciğerde oluşturduğuhistolojik değişiklikler araştırılmıştır.Bu amaçla beta-D-glukan, taksol, taksol+beta-D-glukan ve kontrol grubuolmak üzere 4 grup hayvan kullanılmıştır. Hayvanlar enjeksiyondan önce 24saat süre ile aç bırakılmıştır. Laboratuvar farelerine (Mus musculus) taksol vebeta-D-glukan intraperitonal enjeksiyonla verildikten sonra 6., 12., 24., 48.,saatlerde ve 7., 14., günlerde servikal dislokasyonla öldürülerek karaciğeriçıkarılmıştır.Karaciğerlerin fiksasyonu %10 tamponlanmış nötral formaldehit ileyapıldıktan sonra rutin histolojik işlemler uygulanmış. Preparatlar hematoksilen-eosin, Masson'un üçlü boyaması, periodik asit-schiff (PAS) ve Gomori'ningümüş impregnasyon teknikleri ile boyanmıştır.Çalışmamızdan elde edilen sonuçlara göre; taksolün etkisiylekaraciğerde vakuoler dejenerasyon, sinüzoidlerde genişleme,nekroenflamatuvar odaklar tespit edilmiştir. Sadece beta-D-glukan verilmesininkaraciğerde belirgin bir değişikliğe neden olmadığı görülmüştür. Taksol ve beta-D-glukanın birlikte verildiği gruplarda, taksol gruplarından daha az histolojikdeğişiklik gözlenmiştir. Ayrıca, taksol+beta-D-glukan 14. gün gruplarındakihistolojik görünümün, taksol+beta-D-glukan 7. gün gruplarındaki ilekarşılaştırıldığında normale daha yakın olduğu görülmüştür.Bu bulgular, bir serbest radikal süpürücüsü olan beta-glukanın, taksolünserbest radikal oluşumunu indükleyerek karaciğerde meydana getirdiği oksidatifhasarı azalttığını göstermektedir.Anahtar kelimeler: Taksol, Beta-glukan, Karaciğer, Histolojik Değişiklikler
LIGHT MICROSCOPY INVESTIGATION OF HISTOLOGICAL CHANGES INLIVER INDUCED BY BETA-GLUCAN AND TAXOLIN MICE (Mus musculus)ABSTRACTIn this study, histologic changes induced by taksol which is an anti-neoplastic agent and β-D-glucan which is known as a strong antioxidant andimmunomodulator, were investigated.For this purpose, four groups of animals were used as β-D-glucan, taxol,taxol+ β-D-glucan and control groups. Animals (Mus musculus) were kepthungary for 24 hours before injection, then taxol and β-D-glucan were injectedvia intraperitonally, they were killed by servical dislocation at 6 th, 12 th, 24 th,48 th hours, 7 th, 14 th days and their livers were removed.After isolated livers were fixed in % 10 buffered neutral formalin, routinhistological processes were applied and sections were stained by hematoxilen-eosin, Masson?s trichrome, Periodic asit-Schiff (PAS) and Gomori?s silverimpregnation methods.According to results obtained in our study; histologic changes such asgranular degeneration, dilation at sinuzoids and necroinflumatuar focusesinduced by taxol were determined. It was seen that using only β-D-glucan didn?tcause any obvious differentiation on liver. When taxol and β-D-glucan weregiven together, the degree of histological changes induced by taxol and β-D-glucan, was less than taxol alone. It was seen that the histological picture wasmore close to normal in the taxol+ β-D-glucan 14 th day groups when comparedwith taxol+ β-D-glucan 7 th day groups.These findings show that β-D-glucan, as a free radical sweeper,decreases the oxidative damage that taxol makes in liver by induced freeradical formation.Key Words: Taxol, Beta-glucan, Liver, Histological Changes.
