Abstract
Domates bitkisi, dünyada büyük miktarlarda yetiştiriciliği yapılan bir bitki türüdür. Domatesin besin içeriğinin zenginliği, kullanım alanlarının fazlalığı, tarımının kolay olmasından dolayı üretimi her geçen gün artmaktadır ve dünyada üretimi en çok yapılan yaş sebzedir. Türkiye dünyada domates üretiminde 11,8 milyon ton üretim ile 4. sıradadır. Dünya piyasasında üretim olarak önemli bir yere sahip olan domates aynı zamanda laboratuvar çalışmaları için de model bir bitkidir. Doku kültürü optimizasyonu her genotipe özel olarak yapılmalıdır. Bundan dolayı SC 2121 domates çeşidi için laboratuvarımızda doku kültürü optimizasyon işlemi yapılmıştır. Ticari olarak satın alınan domates tohumu yüzey sterilizasyonu yapılarak, çimlendirilmiştir. Elde edilen steril domateslerin hipokotil ve kotiledonları eksplant kaynağı olarak kullanılmıştır. IAA, NAA, BAP, KN, TDZ, ZEA ve 24-epiBL bitki büyüme düzenleyicilerinin farklı konsantrasyon ve kombinasyonları ile desteklenen MS besiyeri kullanılmıştır. Gelişimi takip edilen eksplantlardan sürgün oluşturanlar alt kültüre alınarak büyütülmüştür. Köklenme ortamında gelişen domatesler saksılara alınarak yeni bitkicikler elde edilmiştir. Yapılan optimizasyon çalışmasının sonucunda sürgün oluşumunda en iyi ortam MS+0,5 mg/L IAA+3 mg/L KN içeren ortam olduğu tespit edilmiştir. Sürgün oluşum yüzdesi, hipokotil %82, kotiledon %78'dir. Kök oluşumunda en iyi ortamlar ise; MS+2 mg/L IAA+0,5 mg/L KN içeren ortam olduğu tespit edilmiştir. Kök oluşum yüzdesi, hipokotil ve kotiledonda %94'tür.
Tomato plant is a plant species grown in large quantities in the world. Because of the richness of the nutrient content of the tomato, the abundance of its uses, the ease of cultivation, its production is increasing day by day, and the most produced age of the world is the vegetable. Turkey is in the 4th place with 11.8 million tons production in tomato production in the world. Tomato, which has an important place in production on the world market, is also a model plant for laboratory studies. Tissue culture optimization should be done specifically for each genotype. Therefore, tissue culture optimization process was performed in our laboratory for SC 2121 tomato variety. Commercially purchased tomatoes are surface-sterilized and germinated. Hypocotyl and cotyledons of the obtained sterile tomatoes were used as explant source. IAA medium was used as MS medium supplement medium supplemented with different concentrations and combinations of NAA, BAP, KN, TDZ, ZEA and 24-epiBL plant growth regulators. Growers were expelled from the explants following the development by subculture. Tomato plants growing in rooting environment were taken in a pot and new plants were obtained. As a result of the optimization study, it was determined that the best environment for shoot formation was medium containing MS + 0.5 mg / L IAA + 3 mg / L KN. The percentage of shoot formation, hypocotyl 82%, cotyledon 78%. The best environments for root formation are; MS + 2 mg / L IAA + 0.5 mg / L KN. The percentage of root formation was 94% in hypocotyl and cotyledon.
