Abstract
Sunulan çalışmada süperior kollikulusdaki toplam nöron sayısı stereolojik metotla odyojenik epilepsi ve kontrol grublarında araştırıldı. İlk olarak bütün sıçanlara odyojenik nöbet testi uygulandı. Ağırlıkları 180-220 gr olan on dişi Wistar sıçan kontrol (n=5) ve epilepsi (n=5) olmak üzere iki gruba ayrıldılar. Sıçanların hepsi üretan (1.25 g/kg; i.p) ile anesteziye alındılar ve sol kardiyak ventrikülden %10'luk formalin ile perfüze edildiler. Kafatası beyinden çıkarıldı ve gruplar sağ sol hemisfer olarak ayrıldı. Beyinlerden 40 μm kalınlığında kesitler alındı ve cresyl violet ile boyandı. Süperior kollikulusun sınırları Paxinos ve Watson'un atlasına uygun olarak belirlendi. Sağ ve sol süperior kollikulusda bulunan toplam nöron sayısı stereolojik optik parçalama metodu kullanılarak hesaplandı. Toplam nöron sayısı, kontrol sağ grubunda 170404,40 ± 14536,414, epileptik sağ grubunda 110335,40 ± 6476,560, kontrol sol grubunda 148328,40 ± 10305,480, epileptik sol grubunda 130103,07 ±5771,297 olarak bulundu. Total nöron sayısı epileptik sağ grubunda kontrol grubuna göre anlamlı ölçüde daha azdı (p<0.05). Kontrol sol ile epileptik sol grubu arasında istatistiksel olarak anlamlı fark yoktu (p>0.05). Epileptik sağ ve epileptik sol grupları arasında toplam nöron sayısında anlamlı fark görüldü (p<0.05). Bununla birlikte kontrol sağ ve kontrol sol grupları arasında fark görülmedi (p>0.05).
In the present study, total number of neurons in the superior colliculus was investigated by a stereologic method in Wistar rats with audiogenic epilepsy and control. Firstly all rats were tested for audiogenic epilepsy. Ten female wistar rats, weighing 180-220 g were divided into two groups: control (n=5) and audiogenic epilepsy (n=5) groups The animals were anesthetized with urethane (1.25 g/kg; i.p.) and perfused through the left cardiac ventricule with 10% formalin.The brains were removed from the cranium and seperated to the right and the left hemispheres in all groups and kept in the same fixative. The brains (right and left hemispheres) were sectioned at 40 μm thickness and stained with cresyl violet. The border of superior colliculus was determined by using a stereotaxic atlas. The total number of neurones were estimated in the right and left superior colliculus in both groups by using stereological optical fractionator method. The total number of neurones in the superior colliculus was 170404,40 ± 14536,414, and 110335,40 ± 6476,560 in the right side and 148328,4 ± 6305,408, and 130103,07 ± 5771,297 in the left side of control and audiogenic groups, respectively. The total number of neurones in the superior colliculus was statistically lower in the right side of epileptic group compared with control (p<0,05). Statistical difference was not observed between left side of epileptic group and control (p>0,05). There was a significant difference between right and left side epileptic group (p<0,05). However, the total number of neurones in the right and left side of control group was not different (p>0,05).
