Publication: Bazı Entomopatojen Fungusların ve Diatom Toprağının Depolanmış Ürün Zararlısı Olan Sitophilus Oryzae (Col.: Curculionidae) ve Tribolium Confusum (Col.: Tenebrionidae) Üzerindeki Etkinliklerinin Belirlenmesi
Abstract
Çalışmada, farklı zararlılardan daha önce elde edilmiş Beauveria bassiana'nın altı farklı izolatı (BÇE 9, BB1/A, BB 1/216, 55-3, BKÜR 1/3A ve BKÜR 1/6B), Isaria fumosorosea (78-3) ve Metarhizium anisopliae (TR-106)' nın birer izolatı, tek başına diatom toprağı ve ikili kombinasyonları önemli bir depo ürün zararlısı olan Sitophilus oryzae (Coleoptera: Curculionidae) ve Tribolium confusum (Coleoptera: Tenebrionidae)' un erginlerine karşı laboratuvar koşullarında etkinlikleri belirlenmiştir. Entomopatojen fungus denemesinde 9 cm çapında Petriler kullanılmış, her bir Petriye 10 adet S. oryzae ve T.confusum ergini konulmuş ve 6 tekerrürlü olarak yürütülmüştür. İzolatların 1 × 108 conidia ml−1 süspansiyonları Petri başına 2 ml olacak şekilde zararlının üzerine direkt uygulanmıştır. Çalışma 21 gün boyunca takip edilerek ölü bireyler not edilmiştir. Diatom toprağı denemesinde ise 180 gr buğday içerisine 125, 250, 500 ve 1000ppm dozlarına karşılık gelecek miktarda diatom toprağı ilave edilerek 3 dakika boyunca çalkalanmıştır. Daha sonra 6 tekerrür olacak şekilde santrifüj tüplerine konulmuştur. Her tür için 10 ergin birey ilave edilmiştir. Deneme de canlı-ölü birey sayımları 7. ve 14.günlerde yapılmıştır. Entomopatojen fungus ve diatom toprağı ikili kombinasyonunda ise 250, 500, 1000 ppm dozundaki diatom toprağı ile 3 fungus izolatının (B.bassianae B:KÜR1/6B, Metarhizium anisopliae TR-106, Isaria fumosorose 78-3) kombinasyonları denenmiştir. Denemede 1 L' lik cam kavanozlara 144 gr buğday alınarak 250, 500, 1000 ppm' e karşılık gelen miktarda diatom toprağı ve kuru olarak Petrilerden elde edilmiş fungus izolatı eklenerek 3 dakika boyunca karıştırılmıştır. Santrifüj tüplerine 4 tekerrür olacak şekilde 36 gramlık buğday alınmıştır. Her bir tüpe 10'ar adet aynı gün çıkış yapmış ergin bireyler ilave edilmiştir. Denemede canlı-ölü birey sayımı 7.gün yapılmıştır. Çalışmaların tümü 24 ℃ ve 28 ℃ olmak üzere iki farklı sıcaklıkta, tamamen karanlık inkibatörde ve % 70 ± 5 bağıl nemde (RH) yürütülmüştür. Denemelerin sonucunda tek başına entomopatojen fungus, tek başına diatom toprağı ve ikili kombinasyonları her iki zararlı mücadelesinde kullanımının uygun olduğu görülmüştür.

In the entomopathogenic fungus experiment, 9 cm diameter Petris were used, 10 S. oryzae and T. confusum adults were placed in each Petri dish and 6 replications were carried out. 1×108 conidia ml-1 suspensions of the isolates were applied directly on the pest at 2 ml per Petri dish. The study was followed for 21 days and dead individuals were noted. In the soil diatom test, an amount of diatom soil corresponding to 125, 250, 500 and 1000 ppm doses was added into 180 g wheat and shaken for 3 minutes. Then it was put into centrifuge tubes in 6 repetitions. 10 adults individuals were added for each species. In the experiment, the live-dead individual counts were made on the 7th and 14th days. Combinations of 250, 500, 1000 ppm soil diatom and 3 fungus isolates (B. bassianae B: KÜR1 / 6B, Metarhizium anisopliae TR-106, Isaria fumosorose 78-3) were tested in a dual combination of entomopathogenic fungus and soil diatom. In the experiment, 144 g of wheat was taken into 1 L glass jars and soil diatom corresponding to 250, 500, 1000 ppm and dry fungus isolate obtained from Petri were mixed for 3 minutes. 36 g of wheat was taken into the centrifuge tubes in 4 repetitions. 10 adult individuals who exited the same day were added to each tube. In the experiment, the live-dead individual count was made on the 7th day. All of the studies were carried out at two different temperatures 24 and 28, in a completely dark incubator and at 70 ± 5% relative humidity (RH). As a result of the experiments, it was seen that the use of entomopathogenic fungus alone, soil diatom alone and their combination of two pests was appropriate for both pest control.
