Abstract
Amaç: Akut miyeloid lösemi (AML) tedavisi güç olan hematolojik bir kanserdir. Kullanılan kemoterapik ilaçlara direnç gelişmesi tedaviyi engellemektedir. AML'de kullanılan sitozin arabinozid (Ara-C) apopitotik etkili bir ilaçtır. Timokinon (TQ) ise antioksidan, antienflamatuvar ve antikansorejen etkili, doğal bir ürün olan çörek otunun ekstraktıdır. Bu çalışmada amaç AML hücre hatlarında kemoterapik ajan olan sitozin arabinozid ve doğal bir antikanserojen olan TQ'nun birlikte ve ayrı ayrı kullanılarak hücre çoğalmasının baskılanıp hücreleri apopitoza götürmek üzerine etkileri araştırılarak sonuçları gözlemlemektir. Materyal ve Metod: Çalışmada Kasumi- 6 AML hazır hücre hatları kullanıldı. Özgün besi ortamına alınan hücrelere 48 ve 72 saatlik süreler için üç ayrı konsantrasyonda sitozin arabinozid ve timokinon uygulandı. Annexın V ve PI boyama sonrası flow sitometride her bir grup için apopitoz, viabilite ve hücre çoğalması değerlendirildi. Bulgular: Çalışmada 48 ve 72 saatlik inkübasyon sürelerinde değerlendirilen her iki grupta da etken maddelerin tek başlarına ve birlikte kullanımları sonucunda apoptozda artış, viabilite de ise azalma olması, hücre çoğalmasının baskılandığını ve istatiksel olarak sonuçların anlamlı (p<0,001) olduğunu göstermiştir. Sonuç: Yaptığımız çalışmamızda TQ ve Ara-C uygulanan her grupta gözlemlediğimiz apoptozun artması, viabilitenin ise azalması bize hücre çoğalmasının baskılandığını göstermiştir. The Cytosine Arabinoside (Ara-C) is used in AML is an apoptotik drog. Timokinon (TQ) is an extract of natural black cumin has antioxidant, anti-inflammatory and anticancer effect. The aim of this study was to investigate the effects of a chemotherapeutic agent Ara-C, and a natural anticancer agent of TQ, on apoptosis and cell proliferation of AML cell lines both alone and in combination. Material and Method: In this study, Kasumi-6 AML cell line was used. The Ara-C and TQ were applied to the cell sat three different concentrations for 48 and 72 hours. After anneksin V and PI staining, apoptosis, viability and cell proliferation were evaluated for each group by flow cytometry. Results: In the study, AML cell lines showed a statistically significant on singleuse of the active substances and their combined use showed increase in apoptosis and decrease in viability in bothgroups at 48 and 72 hour in cubation times (p <0.001). Conclusion: In this study, in each group AML cell line TQ and Ara-C were applied, it was observed that apoptosis was increased and viability decreased and consequently cell proliferation was suppressed.
