Abstract
Pseudomonas viridiflava (Burkholder) Dowson ekonomik öneme sahip birçok bitkide hastalığa neden olan Gram negatif ve pektinolitik bir bakteriyel etmendir. Samsun İli, Türkiye marul üretiminde 5. sırada, kıvırcık marul üretiminde ise ilk sıradır. Bu tez, Samsun İli Çarşamba ovasında marul bitkilerindeki bakteriyel hastalık etmenlerin belirlenmesine yönelik bir çalışma olup, 2016 - 2017 yılları arasında kış döneminde yapılan arazi çalışmalarında bakteriyel hastalık etmenlerinin oluşturduğu belirtilere benzeyen hastalıklı bitki örnekleri toplanmıştır. Tütünde aşırı duyarlılık reaksiyonu gösteren 19 izolattan 18'inin LOPAT, patateste yumuşak çürüklük, patojenisite testleri ile16S rRNA ve rpoD gen sekans analizi soucunda Pseudomonas viridiflava olduğu, 1 izolatın ise biyokimyasal testler ve16S rRNA sekans analizine göre Bacillus pumilus olduğu belirlenmiştir. Bu türlerden Pseudomonas viridiflava Türkiye'de ilk kayıt olarak uluslararası indekse giren dergide yayımlanmış, Bacillus pumilus ise Türkiye'de ve Avrupa'da marul bitkiside ilk kayıt olarak yayım aşamasındadır. Pseudomonas viridiflava (Burkholder) Dowson is a Gram-negative, soil bacterium that is pathogenic to economic plants. Samsun province ranks first in terms of productions of curly lettuce in Turkey. Also the city ranks 5th in term of total productions of lettuce in Turkey. This thesis is a study to determine bacterial disease factors in lettuce plants in Carşamba Plain, Samsun Province. During the winter season between 2016 ve 2017, diseased plant samples were collected which were similar to the symptoms of bacterial disease for the study. Eighteen of nineteen isolates showed hypersensitivity reactions in tobacco, LOPAT, soft rot test in potato ve pathogenicity tests were applied for these 18 isolates. At the same time, 16S rRNA ve rpoD gene sequence analysis were performed. As a result of these analyzes it was found that an isolate was Bacillus pumilus. Other isolates were identified as Pseudomonas viridiflava. Pseudomonas viridiflava isolates TK11, MA14, CD21 as the first record in Turkey has been published in journals in the international index ve Bacillus pumilus isolate CD13 is in the process of publication as the first record in the lettuce plant is in Europe ve Turkey.
