Abstract
Bu çalışmada, kudret narı (Momordica charantia L.) biyoaktif bileşiklerinin ekstraksiyonu için klasik çözücü ekstraksiyonu, sokselet ekstraksiyonu, ultrason destekli ekstraksiyon ve basınç destekli ekstraksiyon yöntemleri uygulanmıştır. Suda ekstraksiyon ve ultrason destekli ekstraksiyonlarda (Çözücü= Su; Etil alkol) toplam fenol miktarı, FRAP ve AE değeri göz önünde bulundurulmuş ve yanıt yüzey yöntemi kullanılarak ekstraksiyon koşulları optimize edilmiştir. İstenilirlik fonksiyonuna göre optimum ekstraksiyon koşulları suda ekstraksiyonda 80 C, 15.44 dakika ve 42.28 mL/g çözücü:materyal oranı, ultrason destekli ekstraksiyonda (Çözücü= Su) 50-60 C, %20 titreşim genliği ve 50 dakika ve ultrason destekli ekstraksiyonda (Çözücü= Etil alkol) 30-40C, %50 etil alkol konsantrasyonu, %20 titreşim genliği ve 10 dak olarak belirlenmiştir. Belirlenen optimum koşullar altında tahmin edilen toplam fenol miktarı, FRAP ve AE değerleri sırasıyla suda ekstraksiyon için 20.45 mg GAE/g, 24.05 µmol Trolox/g ve 2.9×10-3 mL/g, ultrason destekli ekstraksiyon (Çözücü= Su) için 14.08 mg GAE/g, 17.96 µmol Trolox/g ve 1.6×10-3mL/g ve ultrason destekli ekstraksiyon (Çözücü= Etil alkol) için 31.88 mg GAE/g, 17.78 µmol Trolox/g ve 2.0×10-3 mL/g' dır. Ekstrakstların charantin miktarı HPLC yöntemi ile belirlenmiştir. Sonuçlar, en yüksek charantin miktarının çözücü olarak etil alkol kullanılarak gerçekleştirilen ultrason destekli ekstraksiyonda elde edildiğini göstermiştir. Su ve zeytinyağı ile karşılaştırıldığında, en etkili çözücünün etil alkol olduğu tespit edilmiştir
In this study, the following methods were employed for the extraction of bioactive compounds of bitter melon (Momordica charantia L.): classical solvent extraction method (using water and olive oil as solvents), soxhlet extraction, ultrasound-assisted extraction (using water and ethanol as solvents) and pressure-assisted extraction. The total phenolic content, FRAP and AE values were taken into account during water extraction and ultrasound-assisted extraction, and the extraction conditions were optimized by using response surface method. The optimal extraction conditions based on desirability function proved to be 80 °C, 15.44 min and 42.28 mL/g solvent to material ratio for water extraction; 50-60 °C, 20% amplitude and 50 min for ultrasound-assisted extraction (using water as solvent); and 30-40°C, 50% ethanol concentration, 20% amplitude and 10 min for ultrasound-assisted extraction (using ethanol as solvent). Under these optimal conditions, the predicted total phenolic content, FRAP and AE values were as follows, respectively: 20.45 mg GAE/g, 24.05 µmol Trolox/g and 2.9×10-3 mL/g for water extraction; 14.08 mg GAE/g, 17.96 µmol Trolox/g and 1.6×10-3mL/g for ultrasound assisted extraction (using water as solvent); and 31.88 mg GAE/g, 17.78 µmol Trolox/g and 2.0×10-3 mL/g for ultrasound-assisted extraction (using ethanol as solvent).
