Abstract
Bu çalışmada, Ordu Arıcılık Araştırma Enstitüsü tarafından sağlanan iki farklı bal arısı ırkının genotiplerinde daha önce farklı ırklarda çalışılan SNP markörleri ile moleküler tanımlaması ve hastalıklara toleransları incelenmiştir. Bal arısı ırklarından elde edilen genomik DNA'lar Real-Time PCR-HRM ve elektroforez işlemlerine tabi tutulmuş ve SNP yönteminin benzerlik çalışmalarında uygulanabilirliği incelenmiştir. DNA örnekleri bal arılarında kireç hastalığına karşı dayanıklılığı sağlayan genlerin tanımlanmasında kullanılan on adet ve bal arısının genom sekanslaması sonucu elde edilen iki adet toplamda oniki adet SNP primeri ile analiz edilmiştir. SNP analizi sonucunda Kafkas bal arısı ırkında dört adet (AMB-00858574, AMB-01151447, AMB-00631190, AMB-00686140) muğla bal arısı ırkında ise altı adet (AMB-00858574, AMB-00612262, AMB-01151447, AMB-00631190, AMB-00674355, AMB-00686140) primer amplikon oluşturmuştur. Dört adet amplikon bu ırklar için benzer çıkmıştır. PCR-Elektroforez analizi sonucunda Kafkas bal arısı ırkında beş adet (AMB-00858574, AMB-00612262, AMB-01151447, AMB-00631190, AMB-00686140), muğla bal arısı ırkında ise yedi adet (AMB-00858574, AMB-00612262, AMB-01151447, AMB-00631190, AMB-00902548, AMB-00674355, AMB-00686140) amplikon bant oluşturmuştur, dört adet (AMB-00858574, AMB-01151447, AMB-00631190, AMB-00686140) amplikon iki ırkta benzer bant oluşturmuştur. Bu çalışma sonucunda SNP markörlerin Kafkas ve Muğla ırklarında hastalıklara karşı toleransın tanımlanmasında kullanılabilirliği ve bu ırkların tanımlama anahtarı olarak değerlendirilebilirliği ortaya çıkmıştır.
In this Study, Molecular identification and disease tolerance of two different honey bee subspecies' genotypes those were obtained from Ordu Apiculture Research Institute were analyzed with SNP markers those were studied before in different subspecies. Genomic DNA's those obtained from honey bee subspecies were subjected to Real-Time PCR-HRM and electrophoresis processes and usage of SNP markers in similarity studies was researched. DNA samples analysed with ten SNP primers those were used for identification of chalkbrood disease resistance genes and two SNP primers those were obtained from honey bee genom sequencing. Result of SNP analyses, four primers (AMB-00858574, AMB-01151447, AMB-00631190, AMB-00686140) amplified in Caucasian honey bee and six (AMB-00858574, AMB-00612262, AMB-01151447, AMB-00631190, AMB-00674355, AMB-00686140) primers amplified in Muğla honey bee. Four amplicons are similar for this subspecies. Result of electrophoresis analyses, five aplicons (AMB-00858574, AMB-00612262, AMB-01151447, AMB-00631190, AMB-00686140) form band in Caucasian honey bee, seven amplicons (AMB-00858574, AMB-00612262, AMB-01151447, AMB-00631190, AMB-00902548, AMB-00674355, AMB-00686140) form band in Muğla honey bee and four amplicons (AMB-00858574, AMB-01151447, AMB-00631190, AMB-00686140) similar for this subspecies. Result of this study, usage of SNP markers in Caucasian and Muğla honey bee subspecies for identification of disease resistance and evaluability as a identification key for this subspecies was emerged.
