Türkiye'de Yayılış Gösteren Mus L., 1758 (Mammalia:Rodentia) Cinsinin Genetik Varyasyonları

Abstract

TÜRK YE'DE YAYILIŞ GÖSTEREN MUS L., 1758 (MAMMALIA: RODENTIA)C NS N N GENET K VARYASYONLARIÖZETBu çalışmada, Türkiye'nin farklı lokalitelerinden 2000-2005 yılları arasında toplanan47 Mus L., 1758 örneği incelenmiştir. %100'lük etil alkol ve +4oC'de korunan kuyruk veböbreklerden total DNA elde etmek için QIAGEN DNA kiti kullanılmıştır. Daha sonra budokulardan yapılmış ve L 15774 ile H 16498 primerleri kullanılarak ev faresi mitokondrialDNA sekansında 15363-15767 pozisyonlarına denk gelen toplam 405 bazlık (tRNA-Progenine ait 54 baz ve D-loop (kontrol bölgesi) bölgesinin sol tarafına ait 351 baz) mtDNAbölgesi PCR yöntemiyle çoğaltılmış, Aynı primerler sekansların elde edilmesindekullanılmıştır.Çalışma alanında bulunan iki mtDNA tipine dayanılarak, Türkiye'de Mus cinsine aitM. m. domesticus ve M. macedonicus olmak üzere iki taksonun varolduğu belirlenmiştir.Çalışma sonucunda M. m. domesticus'a ait 22 ve M. macedonicus'a ait 17 mtDNA haplotipitespit edilmiştir. GENETIC VARIATIONS IN GENUS OF MUS L., 1758 (MAMMALIA: RODENTIA)DISTRIBUTED IN TURKEYABSTRACTIn this study 47 Mus L., 1758 specimens collected between years 2000-2005 fromdifferent locations in Turkey were analyzed. Tail tips and kidneys preserved in 100% ethanoland maintained at +4oC were used to extract total DNA using QIAGEN DNA kit. A total of405 bp fragment (54 bp of tRNA-Pro gene and 351 bp of left end of the D-loop)corresponding to positions between 15363 and 15767 in mouse mtDNA were amplified usingL 15774 and H 16498 primers in a single PCR reaction. The same primers were used for thesequencing reactions.Two types of mtDNA were found in study areas, suggesting that two taxa of Mus, Musmusculus domesticus Rutty, 1772 and Mus macedonicus Petrov and Ruzíc, 1983, occur inTurkey. 22 M. m. domesticus haplotypes and 17 M. macedonicus haplotypes were detected.These haplotypes were used to construct phylogenic trees using both distance and parsimonyapproaches.Keywords: Mus musculus domesticus, Mus macedonicus, tRNA-Pro gene, D-loop region
