Abstract
I RAT {Wistar strain) SEREBELLUMUNDA PURKİNJE HÜCRELERİ ÜZERİNE KADMİYUM VE FLUNARİZİNİN ETKİLERİ ÖZET Bu çalışmada intrakortikal olarak bregmanın sol lateraline kadmiyum sülfat ve kalsiyum antagonistlerinden flunarizin verildikten sonra sıçan serebellumunda, purkinje hücre sayısındaki değişmeler histolojik olarak değerlendirildi. Bregmanın iki milimetre sol lateraline kadmiyum sülfat (0,0021 mg/kg) verildi. Kontrol grubu hayvanlara aynı hacimde serum fizyolojik uygulandı. Kadmiyum sülfat+flunarizin grubuna ise diğer gruplardan farklı olarak intraperitonal yoldan her gün flunarizin (10 mg/kg) verildi. Her deney grubunda bulunan hayvan bir hafta gözlem altında tutularak yaşatıldı. Bir hafta süreyle intrakardial yoldan nötral formalinle perfüzyona alındı. Serebellumdan elde edilen seri frontal kesitler Thionin ile boyandı. Stereotaksik atlas yardımıyla serebellumun Cms I, Crus II, Lobulus simplex bölgelerinde purkinje hücre sayımı yapıldı. Purkinje hücreleri x400 büyütme ile ışık mikroskobunda, mikroskop alanı kaydırılarak sayıldı. Elde edilen sonuçlara göre purkinje hücre yoğunluğu kontrol grubunda, sağ yarımkürede 20,62±0,68/mm (ortalama+SEM), sol hemisferde 20,56±0,79/mm., kadmiyum grubunda sağ hemisferde 14,23±0,34/mm, sol hemisferde ise 14,17±0,44/mm., kadmiyum+flunarizin grubunda sağ hemisferde 18,46±0,59/mm, sol hemisferde 18,20±0,84/mm olarak tesbit edilmiştir. Gruplar arasındaki fark istatistik açıdan önemli olarak bulunmuştur (p Kalsiyum kanal antagonistlerinden flunarizin, kadmiyumun sebep olduğu hücre ölümünü, kalsiyum kanallarını bloke ederek engellemiştir. II THE EFFECTS OF CADMIUM AND FLUNARIZINE ON THE PURKINJE CELLS IN THE RAT {Wistar strain) CEREBELLUM ABSTRACT In this study, changes in the number of purkinje cells in the rat cerebellum were histologically assessed followig injecting cadmium sulphate and flunarizine of the calcium antagonists intracortically into the left lateral of bregma. Cadmium sulphate (0,0021 mg/kg) was injected into the 2-mm-left lateral of bregma. The same volume of physiological serum was applied to the animals in the control group. Apart from the other groups, flunarizine (10 mg/kg) was intraperitenally injected every day to the cadmium sulphate+flunarizine group. The animals in each group were kept alive and obsorved for a week. They were kept in perfusion for a week by applying neutral formalyne intracardially. A series of frontal sections obtained from the cerebellum were dyed by Thionin. By the help of the stereotaxic atlas, a numeration of purkinje cells was made in the Crus I, Crus II and Lobulus simplex areas of the cerebellum. The d purkinje cells were numerated under the light microscope by aggrandizing 400 times and by siding the microscope cite. In accordance with the results of the numeration, the densities of purkinje cells were found to be 20,62±0,68/mm (mean±SEM) in the right hemisphere and 20,56±0,79/mm in the left hemisphere in the control group, 14,23±0,34/mm in the right hemisphere and 14,17±0,44/mm in the left hemisphere in the cadmium group, and 18,46±0,59/mm in the right hemisphere and 18,20±0,84/mm in the left hemisphere in the cadmium+flunarizyne group.
