Abstract
TÜRKİYE TOPRAKLARINDAN İZOLE EDİLEN NÖTROFİLİK- MEZOFİLİK Streptomyces'LERİN NÜMERİK TAKSONOMİSİ ÖZET Bu çalışmada endüstriyel ve biyoteknolojik açıdan büyük öneme sahip olduğu bilinen Streptomyces'lsrm. Türkiye topraklarındaki çeşitliliğinin ve taksonomik özeliklerinin belirlenmesi amaçlanmıştır. Çalışmamızda 7 farklı coğrafîk bölgeye ait 21 ilden alman toprak örnekleri dilüsyon yöntemiyle seyreltilerek cycloheximide (50 p.g mi'1) ve rifampicin (0.5 ug mi''1) içeren kuru starch- casein agar üzerinde 25° C'de 14 gün inkübe edilerek izolasyonu yapılmıştır. Toplam 800 izolattan saf kültürleri yapılabilen 600 izolat susun ISP3 ve ISP4 Agar'lar üzerinde renk gruplama testleri yapıldı. Oluşturulan 26 renk grubundan seçilen 148 izolat suş test organizması olarak belirlendi. Bu test organizmaları 24 tip örneği ve rastgele seçilen 17 duplikat suş ile birlikte besinsel, biyokimyasal, degredasyon, morfolojik ve fizyolojik testlerden oluşan 120 birim karakter bakımından nümerik taksonomiye göre test edildi. Test sonuçlan X TAXON programında, Ssm - COMPLETE analiz yöntemiyle değerlendirilerek% 80 benzerlik düzeyinde kümeler elde edildi. Duplikat suşlann son veri matriksinden çıkartılmasından sonra kalan 172 suş 12 büyük (7-17), 12 küçük (2-5 suş) ve 1 tek üyeli (SMC) olmak üzere 25 taksonomik gruba ayrılmıştır. 11 NUMERICAL TAXONOMY OF NEUTROPHILIC- MESOPfflLIC Streptomyces ISOLATED FROM TURKEY'S SOIL ABSTRACT In this study, we aimed to determine the variability and taxonomic positions of Streptomyces strains which are known and have an importance as industrially and biotechnologicaly in soil of Turkey. Soil samples taken 21 cities belongs to geographically different 7 regions were examined using dilution plate method, and isolated on surface dried strach- casein agar supplemented with cycloheximide (50 ug mT1) and rifampicin (0.5 ug ml'*1) then incubated at 25° C for 14 days. Availed 600 isolated test strains as pure culture from 800 total stock culture were assigned for colour grouping on ISP3 and ISP4 agars. Selected 148 test strains from 26 colur groups were examined with 24 type strains and randomly selected 17 duplicated strains for 120 unit characters including nutrient, degradation, morphological and pigmentation tests based on numerical taxonomy. Test results were evaluated using Ssm - COMPLETE analyses on the X TAXON programme and the clusters obtained with a 80 % high cophenetic correlation values. Excluding of the duplicated strains from final data matrix including 172 strains were assigned to 25 taxonomic groups which consists of 12 major (7-17 strains), 12 minor (2-5 strains), and a single membered clusters (SMC).
