Abstract
ÖZET Amaç: Varikosel infertilite ilişkisine yönelik çalışmaların çoğu testis üzerine yoğunlaşmıştır. Bu çalışmada erişkin ratlarda deneysel varikoselin epididim morfolojisi üzerine olan etkilerini araştırmak amaçlandı. Gereç ve Yöntem: Yirmi adet Sprague-Dawley rata sol renal venin kısmi obstruksiyonu ile deneysel varikosel oluşturulurken oniki adet rat kontrol, oniki adet rat sham grubunu oluşturdu. Gruplardaki ratlardan yarısı deneyin birinci, kalan yansı ikinci ayında sakrifiye edildi. Epididim testislerden ayrılarak kuru ağırlıkları ölçüldü. PAS boyası ile epididim tübül çaplan ölçülürken, epididim epitelindeki apoptozis TÜNEL yöntemi ile araştınldı. Bulgular: Varikosel grubunun epididim ağırlıklan kontrol ve sham gruplarınkinden anlamlı derecede düşük bulundu (p<0.01). Varikoselli ratların iki ay sonraki sol epididim ağırlığı sağa göre düşüktü (p<0.01). Varikoselli ratlann epididim tübül çaplan kontrol ve sham grubundan daha küçüktü (p<0.001). İkinci ayda varikoselli grubun epididim kaput çaplanndaki azalma anlamlıydı (p<0.001). Ayrıca varikoselli ratların ikinci aydaki sol epididim kauda tübül çaplan sağ tarafa göre daha küçük bulundu (p<0.001). Varikoselli ratlarda epididim epiteli esas hücrelerindeki apoptozis oranı sham ve kontrol grubunda daha fazlaydı. Kaput epididim epitelinde apoptotik hücreler diğer segmentlere göre daha fazla ve yön farkı yoktu. Sonuçlar: Bu çalışmada deneysel varikosel, varikoselli taraf epididim ağırlığını, epididim tübül çapını önemli oranda azaltmakta ve epididim tübül epiteli esas hücrelerinde apoptozise yol açmaktadır. ABSTRACT Objective: Most of the studies investigating the relationship between varicocele and infertility are chiefly focused on the testicles. In this study, the impact of experimental varicocele on the epidydimal morphology is investigated in a model of adult. Material and Method: In twenty Sprague-Dawley rats, experimental varicocele was induced via partial obstruction of the left renal vein. Twelve rats served as controls, and another twelve served as shams. Half of the rats in the groups were sacrified by the end of the first month of the experiment, and the rest were sacrified by the end of the second month. The epydidymides were disected off the testicles and their dry weights were measured. Epidydimal tubule diameters were measured via PAS staining and TUNEL assay was used to assess the epithelial apoptosis within the tubules. Findings: Weights of the epididymides in the trial group was found to be significantly lower than those in the control and sham groups (p<0.01). In rats with varicocele, left epididymides weighted significantly lower than the right ones (p<0.01). Epididymal tubule diameters in rats with varicocele were significantly narrower than those in the control and sham groups (p<0.001). The decrease in epidiymal caput diameters in rats with varicocele by the end of the second month was significant (pO.001). At the same time, cauda tubule diameters of the same epididymides were significantly narrower than those on the left as well (p<0.001). Rats with varicocele displayed a significantly increased rate of apoptosis when compared with controls and shams. On the other hand, apoptotic activity within the caudal segment did not display any differences than of the other segments. Conclusions: The findings of this study demonstrate that, experimentally induced varicocele significantly decreases epidydimal weight and tubule diameter on the involved side and provoke apoptosis within the proper epithelial cells. Experimental varicocele inflicts damage to the opposite epidydimes as well, and this damage increases over time. Key words: epididymis, experimental varicocele, apoptosis, adult rat VII
