Publication: Organik Tavuk Parça Etlerinde Salmonella Serotipleri İle İzolatlarda Antibiyotik Direnç Profilinin Belirlenmesi
Abstract
Amaç: Bu çalışmada, Samsun ilinden temin edilen organik tavuk parça etlerinde Salmonella spp. varlığının, klasik kültür ve IMS teknikleri kullanılarak araştırılması, elde edilen izolatların oriC gen sekansı esas alınarak PCR ile doğrulanması ve izolatların serotiplerinin belirlenmesi ile antibiyotiklere karşı dirençlilik durumlarının saptanması amaçlanmıştır. Materyal ve Metot: Mayıs-Ağustos 2015 tarihleri arasında 4 ay süre ile paketlenmiş formdaki toplam 150 organik tavuk parça etinde (50 but, 50 kanat, 50 derisiz- göğüs eti) analiz edildi. Örneklerdeki Salmonella klasik kültür ve IMS tekniği ile izole edildi. Şüpheli izolatlar MALDI-TOF yöntemi ile Salmonella spp. olarak belirlendi. PCR yöntemiyle izolatların konfirmasyonu tamamlandı. İzolatların antibiyotiklere karşı dirençliliği VITEK 2 Compact Technology yöntemi ile saptandı. Serolojik olarak Salmonella serotipleri belirlendi. Bulgular: İncelenen 150 örnekten 42'sinin (%28) Salmonella spp. ile kontamine olduğu saptandı. Yapılan serotiplendirmede 93 izolatın 53'ü (%56.9) Salmonella enterica serotip Infantis, 40'ının (%43.1) ise Salmonella enterica serotip Anatum olduğu belirlendi. Antibiyotik direnç profili analizi sonucu S. Infantis izolatlarının tamamı, sefuroksim aksetile, amikasine, gentamisine ve siprofloksasine karşı dirençli bulunurken, izolatlardan 1'i ampisiline, 16'sı trimetoprim\sülfametaksazole, 1'i ise seforoksime karşı dirençli bulunmuştur. Benzer şekilde S. Anatum izolatlarının tamamı amikasine, gentamisine ve siprofloksasine karşı dirençli bulunurken, izolatlardan 2'si trimetoprim\sülfametaksazole karşı dirençli bulunmuştur. Sonuç: Organik tavuk etinde Salmonella spp. elde edilmesi ve izolatların antibiyotiklere direnç göstermesi halk sağlığı açısından oldukça önemlidir. Aim: The aim of this study is to examine the presence of Salmonella spp. in organic chicken parts supplied from Samsun province by using classical culture and IMS techniques; to verify the obtained isolates with PCR based on oriC gene sequence and to determine the serotypes of isolates and to find out their resistance against antibiotics. Material and Method: 150 pieces of organic chicken parts (50 drumstick, 50 wings, 50 chicken breast without skin) in packaged form were analyzed for four months between May and August 2015. Salmonella in the samples was isolated with classical culture and IMS technique. Suspicious isolates were determined as Salmonella with MALDITOF method. Isolates were confirmed with PCR method. The resistance of the isolates against antibiotics were found with VITEK 2 Compact Technology method. Serotypes of Salmonella were determined serologically. Results: Of the 150 samples analyzed, 42 (28%) were found to be contaminated with Salmonella spp. In the serotyping performed, 53 (56.9%) out of 93 isolates were found to be Salmonella enterica serotip Infantis, while 40 (43.1%) were found to be Salmonella enterica serotip Anatum. According to antibiotic resistance profile analysis results, while all of the S. Infantis isolates were found to be resistant to cefuroxime axetil, amikacin, gentamicin and ciprofloxacine; 1 was found to be resistant to ampiciline, 16 were found to be resistant to trimethoprim\sulfamethoxazole and 1 was found to be resistant to cefuroxime. Similarly, while all of the S. Anatum isolates were found to be resistant to amikacin, gentamicin and ciprofloxacine, 2 of the isolates were found to be resistant to trimethoprim\sulfamethoxazole. Conclusion: Salmonella spp. in organic chicken meat and the resistance of the isolates to antibiotics is very important for public health.
