ÖZETSAMSUN İLİ BAFRA VE ÇARŞAMBA İLÇELERİ SERALARINDAKİ KÖK-UR NEMATODLARI (NEMATODA: MELOIDOGYNIDAE: MELOIDOGYNESPP.) TÜR VE IRKLARININ TESPİTİ İLE YAYILIŞ VE BULAŞIKLIKORANLARI ÜZERİNDE ARAŞTIRMALARBu çalışmanın amacı, Samsun İli Bafra ve Çarşamba ilçeleri seralarındabulunan kök-ur nematodlarının yayılış ve bulaşıklık oranları ile tür ve ırklarınınsaptanmasıdır.Araştırmanın materyalini, Bafra ve Çarşamba ilçeleri seralarından getirilen, kök-urnematodu ile bulaşık bitki materyali ve topraklar oluşturmuştur. Bafra ilçesinde 10köydeki 50 seradan, Çarşamba ilçesinde ise 6 köydeki 100 seradan, toprak ve kökörnekleri alınıp, kökler 0-4 bulaşıklık ıskalasına göre değerlendirilmiştir.Yayılış ve bulaşıklık tespiti çalışmaları neticesinde; bulaşıklığın Bafra ilçesiseralarında % 52, Çarşamba'da ise % 64 oranlarında olduğu saptanmıştır. Ayrıcabulaşıklık seviyesi seralardaki sebze çeşitlerine göre değerlendirildiğinde, zararın enfazla hıyar (% 51,1) ve domates bitkisinde (% 34,5) olduğu belirlenmiştir.Kök-ur nematodlarının tür teşhisinde, araziden getirilen bulaşık materyaller?Blender-Elek? metoduna tabi tutularak elde edilen dişilerin, gliserin ortamındahazırlanan perineal preparatlarından yararlanılmıştır.Tür teşhisleri çalışmaları neticesinde; en yaygın türün % 45,6 oranı ile M. incognita(Kofoid, White 1919) Chitwood 1949 olduğu, bu türü % 26,2 oranı ile M. arenaria(Neal, 1889) Chitwood 1949, % 23,5 oranı ile M. hapla Chitwood 1949 ve % 4,7 oranıile M. javanica (Treub, 1885) Chitwood 1949'nın izlediği saptanmıştır.Tür teşhisi sonucunda, tek tür olduğu belirlenen populasyonların saf kültürlerindenelde edilen yumurtalar steril topraklardaki domates fidelerine verilerek, 30 gün sonundafideler sökülmüş ve her populasyon için elde edilen yumurtalar ?Kuzey KarolinaKonukçu Testi? için kullanılarak ırklar belirlenmiştir.Bafra ilçesinde M. incognita populasyonlarının % 62,5'i M. incognita ırk 2, %37,5'i M. incognita ırk 4, M. arenaria populasyonlarının % 50'si M. arenaria ırk 1, %50'si ise M. arenaria ırk 2 olarak saptanmıştır. Çarşamba ilçesinde ise M. incognitapopulasyonlarının % 80'inin M. incognita ırk 2, % 20'sinin M. incognita ırk 4 olduğubelirlenmiştir.Anahtar Kelimeler : Kök-ur nematodları, sera, yayılış, bulaşıklık, tür, ırk.
ABSTRACTRESEARCH ON DISTRIBUTION AND INFESTATION RATIO WITHSPECIES AND RACE DETERMINATION OF ROOT-KNOT NEMATODES(NEMATODA: MELOIDOGYNIDAE: MELOIDOGYNE SPP.)OF GREENHOUSES IN BAFRA AND ÇARŞAMBA PROVINCE IN SAMSUNThe aim of the study was the determination of species and races with distributionand infestation rates of root-knot nematodes found in greenhouses in Bafra andÇarşamba.Plant and infested soil collected from Bafra and Çarşamba grenhouses were used asa research materials. Soil and root samples were taken from 50 greenhouses in 10villages on Bafra plain and 100 greenhouses in 6 villages on Çarşamba plain. Rootswere evaluated according to 0-4 scale.As a result of distribution and infestation studies, it was found that 52 % in Bafraplain greenhouses and 64 % in Çarşamba greenhouses were infected with root-knotnematodes. The infestation rate was also evaluated according to vegetable variety ingreenhouses. Cucumbers (51,1 %) and tomatoes (34,5 %) were infected at the highestlevels.Female nematodes were extracted by using ?Blender-Sieve? combined methods andperineal patterns mounted in glycerol were used for species identification.M. incognita (% 45,6) was the dominant species, followed by M. arenaria (%26,2), M. hapla (% 23,5) and M. hapla (% 4,7).After the species identification, eggs of populations from single species wereintroduced to tomato seedlings in sterilized soils. The seedlings were uprooted 30 dayslater for checking. An eggs from each population were used for ?North Carolina HostTest? in order to determine races. M. incognita race 2 was 62,5 %, M. incognita race 4was 37,5 %, M. arenaria race 1 was 50 % and M. arenaria race 2 was 50 % on Bafraplain.
