Publication: Domates Genotiplerinde Genetik Benzerliğin SSR (Basit Dizi Tekrarı) Marköriyle Tespiti
Abstract
Bu araştırma, domates (Solanum lycopersicum) popülasyonlarında genetik çeşitliliği belirlemek amacıyla farklı bölgelerden elde edilen 21 yerli ve 3 ticari genotipin 40 SSR lokusu kullanılarak genetik düzeyde allel profilleri, genetik tanımlamaları ve aralarındaki genetik benzerlikleri araştırılmıştır. Lokuslarda gözlenen allel sayısı 13 (LEat018) ilâ 3 arasında (SSR50, SSR80, SSR9, LEtat002) değişmiştir. Beklenen ve gözlenen heterozigotluk oranı ortalaması sırasıyla He: 0,753 ve Ho:0,714 olarak bulunmuştur. Lokuslara ait ortalama Polimorfizm Bilgi İçeriği (PIC) değeri, 0,694 olarak bulunmuştur. Allellerin lokuslarda ki dağılımlarına bakıldığında; en yüksek allel frekansının % 62,5 ile SSR75 lokusunda, en düşük allel frekanslarının ise % 16,6 ile AI491065 ve LEta024 lokuslarında görüldüğü tespit edilmiştir. Kümeleme analizi için UPGMA (Unweighted Pair-Group Method Using Arithmetic Means) yöntemi uygulanmıştır. Dendogramda 2 ana grup ortaya çıkarak, genotiplerin büyük kısmını içeren ikinci (2) grup içinde çok sayıda alt grup gözlenmiştir. 40 SSR lokusuna göre analiz edilen 24 domates genotipinden Bafra-Merkez ve Sivas-Yukarıkale Köyü genotiplerinin genetik benzerlik değeri 0,776 olarak bulunmuştur. En düşük benzerlik 0,136 değeri ile Sivas-Yıldızeli-Emirler Köyü ve Samsun- Çarşamba genotipleri arasında bulunmuştur.
In this research, allele profiles, genetic identification and genetic similarity between 21 local 3 commercial genotypes collected from different regions were investigated to establishe the genetic variability in tomatoes (Solanum lycopersicum) populations at genetic level by using 40 SSR loci. Number of alleles per locus ranged from 13 (LEat018) to 3 (SSR50, SSR80, SSR9, LEtat002). Expected and observed heterozygosity values were between He: 0,753 and Ho: 0,714 respectively. The mean Polymorphism Information Content (PIC) value of these loci was found as 0,694. Considering distribution of alleles per loci, the highest allele frequency was observed from SSR75 with % 62,5 while the lowest was in AI491065 and LEta024 with % 16,6 locus. The dendrogram was consisted 2 main groups, and group 2 was the largest and it contained many sub-groups.
