This chapter provides information about stereology in general and physical and optical disectors in particular. The anatomy and histology of the rat cerebellum are also briefly described. We also outline the procedures involved in stereological study of the cerebellum, explaining the tissue processing and staining methods required. Furthermore, this chapter discusses the application of the physical disector for estimating the numerical density of Purkinje cells and of Cavalieri's principle for estimating cerebellar volume. The total number of Purkinje cells was estimated using these procedures. The coefficient of error (CE) and the coefficient of variance (CV) values required for ensuring the precision of the stereological design are additionally explained in detail. © 2024 Nova Science Publishers, Inc.
